Loss of PKM2 dysregulates inflammatory signaling in the infarcted murine heart.

Inflammation and a metabolic shift from oxidative metabolism to glycolysis are common in the ischemic heart, the latter partly controlled by pyruvate kinase (muscle, PKM). We previously identified alternative splicing promoting the PKM2 isoform after myocardial infarction (MI). We examined the role of PKM2 physiological upregulation after MI, modeled by ligation of the left anterior descending coronary artery, using global PKM2 knockout (PKM2) mice.

IL-35 May Prevent the Exacerbation of Aspiration Pneumonia Involving Porphyromonas gingivalis by Suppressing IL-17 Production.

Periodontitis was reported to be associated with aspiration pneumonia. However, the relationship between periodontitis and aspiration pneumonia remains unclear. This study investigated the virulence factor of Porphyromonas gingivalis, which exacerbates aspiration pneumonia, and the role of IL-35, an inhibitory heterodimeric cytokine of EBI3 and p35, in aspiration pneumonia using Ebi3 knockout (KO) mice.

Photodynamic disruption of a polymicrobial biofilm of two periodontal species using indocyanine green-loaded nanospheres.

Objective: Antimicrobial photodynamic therapy (aPDT) is considered a potential treatment for biofilm infections, which have become an increasing health issue because of the rise in antimicrobial resistance. This study aimed to evaluate the bactericidal effect of aPDT using indocyanine green-loaded nanospheres with chitosan coating (ICG-Nano/c) against polymicrobial periodontal biofilms.

Methods: Composite biofilms of Porphyromonas gingivalis and Streptococcus gordonii were constructed in 96-well plates, and aPDT with ICG-Nano/c and an 810 nm diode laser was performed either by direct irradiation or transmitting irradiation through a 3-mm-thick gingival model.

Glycocalyx Interactions Modulate the Cellular Uptake of Albumin-Coated Nanoparticles.

Albumin-based nanoparticles (ABNPs) represent promising drug carriers in nanomedicine due to their versatility and biocompatibility, but optimizing their effectiveness in drug delivery requires understanding their interactions with and uptake by cells. Notably, albumin interacts with the cellular glycocalyx, a phenomenon particularly studied in endothelial cells. This observation suggests that the glycocalyx could modulate ABNP uptake and therapeutic efficacy, although this possibility remains unrecognized.
