Article Synopsis
  • A phase II trial investigated the effectiveness of combining enzalutamide with fulvestrant in treating ER+/HER2- breast cancer to see if it reduces residual tumors at surgery compared to fulvestrant alone.
  • * The combination treatment showed a significantly higher rate of achieving complete tumor response (PEPI=0) and improved immune activation in tumors compared to fulvestrant only.
  • * Results indicated that the combination therapy not only lowered Ki67 levels (a marker for cancer proliferation) but also altered the tumor immune environment, suggesting increased anti-tumor activity.

Background: Advances in technology and knowledge have facilitated both an increase in the number of patient variants reported and variants reclassified. While there is currently no duty to recontact for reclassified genetic variants, there may be a responsibility. The purpose of this clinical practice advisory document is to provide healthcare practitioners guidance for recontact of previously identified and classified variants, suggest methods for recontact, and principles to consider, taking account patient safety, feasibility, ethical considerations, health service capacity and resource constraints.

Article Synopsis
  • TDO2 is expressed at higher levels than IDO1 in triple-negative breast cancer (TNBC).
  • Knocking down TDO2 can trigger an increase in IDO1 as a compensatory response.
  • A new dual inhibitor targeting both TDO2 and IDO1 reduces tryptophan catabolism, inhibits anchorage-independent survival, and decreases invasive capabilities of cancer cells.

Ovarian cancer is the deadliest gynecologic malignancy, and therapeutic options and mortality rates over the last three decades have largely not changed. Recent studies indicate that the composition of the tumor immune microenvironment (TIME) influences patient outcomes. To improve spatial understanding of the TIME, we performed multiplexed ion beam imaging on 83 human high-grade serous carcinoma tumor samples, identifying approximately 160,000 cells across 23 cell types.

The term "recurrent constellations of embryonic malformations" (RCEM) is used to describe a number of multiple malformation associations that affect three or more body structures. The causes of these disorders are currently unknown, and no diagnostic marker has been identified. Consequently, providing a definitive diagnosis in suspected individuals is challenging.
