An indirect detection method using high-performance immunoaffinity chromatography (HPIAC) was used to measure low levels of an analogue of bovine Growth Hormone Releasing Factor (bGHRF). An antibody (Ab) labelled with alkaline phosphate (ALP) was incubated with the bGHRF analogue to perform a complex between the antigen (Ag) and the antibody-enzyme (Ab-En) conjugate. The complex was then injected onto a cartridge containing an immobilized Ag affinity support.

We have developed high-performance immunoaffinity chromatography (HPIAC) methods for the detection and quantitation of bovine growth hormone releasing factor (GHRF), which could also be applicable to its metabolites in biofluids. These approaches have involved a combination of IAC using immobilized antibody (Ab) to GHRF, together with reversed-phase high-performance liquid chromatography (RP-HPLC) separations of initially isolated and concentrated protein, followed by selective detection, involving on-line immunodetection (ID) schemes. ID methods involved HPIAC supports of the Ab, together with synthesized Ab-fluorescein isothiocyanate conjugates.

Eighteen normal cats were randomly allocated into two blocks with three treatment groups and dosed orally with clindamycin aqueous solution for 10 days at a dosage rate of 5.5 mg/kg twice daily (Group 1), 11 mg/kg twice daily (Group 2), or 22 mg/kg once daily (Group 3). At the end of dosing, all cats were killed and tissues were taken for clindamycin concentration analysis.

Four multiparous Holstein cows were utilized to determine the effects of high doses of recombinant bovine somatotropin (rbSt) on milk production, feed intake, somatotropin concentration in milk, blood and urine constituents and the presence or absence of treatment-related lesions. Treatments were 0 (control) or 430 mg rbSt daily for 21 days as im injections beginning approximately 150 days into lactation. Cows were fed concentrate according to level of milk production and corn silage ad libitum.
