Article Synopsis
  • - Viremic non-progressors (VNPs) are a rare group of HIV-1 individuals who maintain normal CD4 T cell counts despite high viral loads, resembling natural hosts of simian immunodeficiency virus, but the reasons for this are not fully understood.
  • - A study using single-cell and multiomics methods examined 16 VNPs and 29 HIV+ progressors, revealing genetic factors like CCR5Δ32 heterozygosity and lower CCR5 expression, alongside reduced intestinal disruption and immune responses in VNPs.
  • - The research highlights various traits contributing to the immune stability in VNPs, indicating important insights for potential HIV treatment strategies in the future.

Theropithecus gelada, the last surviving species of this genus, occupy a unique and highly specialised ecological niche in the Ethiopian highlands. A subdivision into three geographically defined populations (Northern, Central and Southern) has been tentatively proposed for this species on the basis of genetic analyses, but genomic data have been investigated only for two of these groups (Northern and Central). Here we combined newly generated whole genome sequences of individuals sampled from the population living south of the East Africa Great Rift Valley with available data from the other two gelada populations to reconstruct the evolutionary history of the species.

Article Synopsis
  • Two new diagnostic classifications for acute myeloid leukemia (AML) were introduced in 2022 to enhance understanding of the disease's biology compared to the previous 2017 categories.
  • A study involving whole exome sequencing on 69 patients with intermediate-risk AML revealed that 45% had their diagnostic category changed, with a notable increase in those classified as AML with myelodysplasia-related changes due to a high mutation frequency.
  • The findings highlighted that specific gene mutations, particularly in the most commonly mutated gene, correlated well with myelodysplastic syndrome history, while also indicating that some newly diagnosed patients without prior dysplasia had these mutations, suggesting the need for a more unified classification system.

Noble element time projection chambers are a leading technology for rare event detection in physics, such as for dark matter and neutrinoless double beta decay searches. Time projection chambers typically assign event position in the drift direction using the relative timing of prompt scintillation and delayed charge collection signals, allowing for reconstruction of an absolute position in the drift direction. In this paper, alternate methods for assigning event drift distance via quantification of electron diffusion in a pure high pressure xenon gas time projection chamber are explored.

Somatic single-nucleotide variants (SNVs) occur every time a cell divides, appearing even in healthy tissues at low frequencies. These mutations may accumulate as neutral variants during aging, or eventually, promote the development of neoplasia. Here, we present the SP-ddPCR, a droplet digital PCR (ddPCR) based approach that utilizes customized SuperSelective primers aiming at quantifying the proportion of rare SNVs.
