Stability enhancement of Amphotericin B using 3D printed biomimetic polymeric corneal patch to treat fungal infections.

Amphotericin B eye drops (reconstituted from lyophilized Amphotericin B formulation indicated for intravenous use) is used off-label for fungal keratitis. However, the reconstituted formulation is stable only for a week, even after refrigeration. Moreover, a high dosing frequency makes it an inconvenient treatment practice.

Ocular delivery of Amphotericin B: current challenges and future perspectives.

Introduction: Amphotericin B is a polyene antibiotic that is used as an off-label eye drop to treat fungal keratitis. Poor solubility, permeability and high susceptibility to hydrolytic degradation make it challenging to formulate a drug delivery system. Methods: EVs were isolated from vitreous of patients with bacterial endophthalmitis (culture positive and culture negative) and noninfectious control by exosome isolation reagent and characterized, and the levels of C8α and calpain-2 was assessed by enzyme-linked immunosorbent assay in isolated EVs and direct vitreous.
