Regulators of chromatin accessibility play key roles in cell fate transitions, triggering onset of novel transcription programs as cells differentiate. In the male germ line stem cell lineage, tMAC, a master regulator of spermatocyte differentiation that binds thousands of loci, is required for local opening of chromatin, allowing activation of spermatocyte-specific promoters. Here we show that a cell-type specific surveillance system involving the multiple zinc finger protein Kmg and the pipsqueak domain protein Dany dampens transcriptional output from weak tMAC dependent promoters and blocks tMAC binding at thousands of additional cryptic promoters, thus preventing massive expression of aberrant protein-coding transcripts.

For a new clinical trial testing a group retreat-based format of psilocybin-assisted therapy, our research team created an initial set of practice guidelines that aimed to describe facilitator use of touch in a way that is ethical, supportive, and minimizes harm. In our first three retreats, however, we had two unexpected experiences with touch that led us to iterate our initial guidelines into a new version of guidelines. In this Technical Report, we describe our evolving guidelines specifying acceptable practices for facilitator use of touch to ensure a safe, supportive, and therapeutic participant experience.

Purpose: Non-Hispanic Black (NHB) women in the United States exhibit higher prevalence and mortality rates from cardiovascular diseases compared to non-Hispanic White (NHW) women. Previous studies in NHB men have demonstrated impaired vascular function at rest and during exercise; however, to date, no studies have investigated the hyperemic responses during exercise in NHB women. Therefore, we tested the hypothesis that, compared with NHW women, NHB women would present an attenuated increase in forearm blood flow (FBF) and forearm vascular conductance (FVC) during steady-state rhythmic handgrip exercise.
