Article Synopsis
  • Chronic anterior uveitis (CAU) can cause serious eye problems, so doctors created treatment plans to help kids with it.
  • The study looked at how these treatment plans worked for kids with a type of arthritis-related eye condition, tracking their medicine and eye health over time.
  • Results showed that most kids had good control of their uveitis after six months, and both treatment plans were successfully used in real-life doctor visits.

Article Synopsis
  • The study aimed to investigate if patients with systemic juvenile idiopathic arthritis (sJIA) have a higher occurrence of rare genetic variants associated with hemophagocytic lymphohistiocytosis (HLH) compared to controls.
  • Researchers performed targeted sequencing on key HLH-related genes in sJIA patients and compared results against a large control group, finding significant associations with rare variants, particularly in the STXBP2 and UNC13D genes.
  • The results suggest that rare HLH variants may play a role in the development of sJIA, with distinct patterns observed in patients with and without macrophage activation syndrome (MAS), indicating a complex genetic influence on the condition.

Article Synopsis
  • The study evaluates the safety, tolerability, and effectiveness of tofacitinib in children and adolescents with juvenile idiopathic arthritis (JIA) in a long-term extension (LTE) study.
  • Out of 225 patients treated with tofacitinib, a high percentage experienced adverse events (AEs), although serious AEs were relatively low, with a focus on infection rates and disease activity.
  • Results showed significant improvement in disease activity scores over time, with a notable increase in patients achieving inactive disease status after 48 months of treatment.

Alcohol use and anxiety disorders occur in both males and females, but despite sharing similar presentation and classical symptoms, the prevalence of alcohol use disorder (AUD) is lower in females. While anxiety is a symptom and comorbidity shared by both sexes, the common underlying mechanism that leads to AUD and the subsequent development of anxiety is still understudied. Using a rodent model of adolescent intermittent ethanol (AIE) exposure in both sexes, we investigated the epigenetic mechanism mediated by enhancer of zeste 2 (EZH2), a histone methyltransferase, in regulating both the expression of activity-regulated cytoskeleton-associated protein (Arc) and an anxiety-like phenotype in adulthood.

Objective: To evaluate whether there is an enrichment of rare variants in familial hemophagocytic lymphohistiocytosis (HLH) genes and systemic juvenile idiopathic arthritis (sJIA) with or without macrophage activation syndrome (MAS).

Methods: Targeted sequencing of HLH genes (, , , , , ) was performed in sJIA subjects from an established cohort. Sequence data from control subjects were obtained (dbGaP:phs000280.
