We describe a new assay for the chlorination activity of myeloperoxidase and detection of chloramines. Chloramines were detected by using iodide to catalyze the oxidation of either 3,3',5,5'-tetramethylbenzidine (TMB) or dihydrorhodamine to form strongly absorbing or fluorescent products, respectively. With TMB as little as 1 muM taurine chloramine could be detected.
View Article and Find Full Text PDFThe present report concerns the generation of specific markers and the establishment of a selection procedure for microglia specific molecules from phage displayed peptide libraries. Negative selection against a mouse monocytic cell line (IC-21) and positive selection against primary mouse microglia was combined in the selection procedures using a mixture of two random peptide libraries displayed on phage. In a first set of experiments, one clone was selected that bound microglia and IC-21 cells to equal extent, and three clones that bound to unsorted primary microglia to substantially higher levels than to IC-21 cells.
View Article and Find Full Text PDFThe synthesis of a series of neoglycolipid conjugates of foscarnet as potential drug targeting forms or lipophilic prodrugs of foscarnet is described. The compounds were obtained from suitably protected neoglycolipids, in which the lipid chain consisted of 12 to 20 carbon atoms, by ethoxycarbonylphosphonylation at the 6-hydroxyl or 4-hydroxyl group followed by deprotection. The in vitro antiviral activity of the compounds was determined in human foetal lung cells infected with human cytomegalovirus (HCMV) or herpes simplex virus type 1 (HSV-1).

Since specific blockers of the N-methyl-D-aspartate (NMDA) receptor channel prevented both types of cell death, the role of Ca2+-dependent processes in the initiation of glutamate toxicity was further investigated. We examined the possible involvement of mitochondria and the role of the Ca2+/calmodulin-regulated protein phosphatase, calcineurin, in the development of either type of cell death.

This resulted in high expression (0.7% of the microsomal protein).
