The cis-regulatory elements encoded in an mRNA determine its stability and translational output. While there has been a considerable effort to understand the factors driving mRNA stability, the regulatory frameworks governing translational control remain more elusive. We have developed a novel massively parallel reporter assay (MPRA) to measure mRNA translation, named Nascent Peptide Translating Ribosome Affinity Purification (NaP-TRAP).

Article Synopsis
  • Mutations causing premature termination codons (PTCs) in protein-coding genes lead to severe, often life-threatening genetic diseases that currently lack approved treatments.
  • Scientists are exploring suppressor tRNAs (sup-tRNAs) that could potentially translate these PTCs and restore protein synthesis, but developing efficient and specific sup-tRNAs is challenging.
  • This research introduces a new approach using a naturally occurring pyrrolysine tRNA (tRNAPyl) to create a series of engineered suppressor tRNAs (PASS-tRNAs), which successfully restored protein synthesis in both bacterial and human cells, showing promise for treating genetic disorders like BRCA1 mutations in cancer.

Article Synopsis
  • Bicuspid aortic valve (BAV) shows increased progression of aorta dilation compared to tricuspid aortic valve (TAV) in patients with aortic stenosis, with a median annualized change of 0.33 mm/year for BAV versus 0.21 mm/year for TAV.
  • Factors influencing the dilation rate differ by valve type; BAV patients are more affected by low-density lipoprotein (LDL) levels, while TAV patients are influenced by the apolipoprotein B/A-I ratio and baseline aortic jet velocity.
  • The study highlights that men and women have different predictors for AA dilation; men’s dilation relates to baseline jet velocity and aortic diameter, while

  • Long-acting reversible contraceptives (LARCs) are highly effective and essential for empowering patients' reproductive choices, prompting a quality improvement (QI) initiative to enhance access at a health center from March to June 2019.
  • An evaluation study used the Wilcoxon-rank test to analyze the impact of the QI interventions on the delivery of LARCs and related reimbursements.
  • Results showed a significant increase in LARC deliveries and an approximate $1,000 monthly rise in reimbursements post-intervention, suggesting successful QI implementation but highlighting the need for further research on equitable access across different populations.

Article Synopsis
  • * A study involving blood and urine samples from both ADPKD patients and healthy volunteers revealed significant increases in toxic bile acids among the ADPKD group.
  • * The results suggest that elevated bile acids and specific compounds may heighten the risk of drug-induced liver injury and drug interactions in ADPKD patients due to impaired liver transport mechanisms.
