The aim of the study was assessment of effectiveness and safety of extracorporeal procedures (plasmapheresis) in prevention and combined treatment of acute rejection crises (ARC) after kidney transplantation in patients at high risk to develop acute immunological conflicts in early postoperative period. 94 patients aged 21-56 years after allotransplantation of cadaver kidney (ATCK) entered the study. They were divided into two groups: group 1 patients (n = 47) received plasmapheresis procedures in combined preventive and therapeutic treatment of ARC; matched group 2 patients (n = 47, retrospective) received the same therapy but plasmapheresis.

The Tth DNA polymerase genes from the thermophilic bacterium Thermus thermophilus (strans HB-8 and Tt-111) were cloned and expressed in Escherichia coli cells. The target protein contained a polyhistidine tag at the N-terminus that allowed its single-stage isolation by affinity chromatography on Ni-NTA-agarose. The isolated enzyme displayed both high DNA polymerase and reverse transcriptase activities.

Plasmids carrying an asd gene from a mutant. S-(2-aminoaethyl)-L-cysteine resistant strain of Corynebacterium glutamicum were selected from a clonoteque constructed on a plasmid cloning vector pSL5 by complementation of asd mutation in Escherichia coli. The histidine operon of Salmonella typhimurium and its fragments were cloned in Escherichia coli cells on a multicopy plasmid. Expression of the cloned genes and histidine production by the variants possessing the hisG mutation which desensibilizes the ATP phosphoribosyl transferase for histidine were studied. Amplification of the complete operon including the hisG gene enables histidine accumulation of 2-3 g/l after 72 hours of fermentation.

Expression of the histidine operon in Escherichia coli cells in contrast to the one in Salmonella typhimurium is changed proportionally to cells growth rate on the different carbon sources. The specific activity of histidinol-dehydrogenase is repressed by addition of 19 amino acids both in Escherichia coli and Salmonella typhimurium independent of the growth medium used. Using of Escherichia coli and Salmonella typhimurium strains containing the heterologous histidine operons made possible to demonstrate the dependence of the histidine operon metabolic regulation to be determined by the operon itself but not by the specificity of the recipient cells.

The effect of active arterial-hypertension preventive programs on the epidemiologic situation was studied over a nine-year period, 1977 through 1985. In the first five years, the number of effectively treated cases increased, while essential hypertension morbidity associated with temporary disability diminished, as did mortality and the incidence of myocardial infarction and cerebral accidents among 40- to 54-year-old male industrial workers. After an active preventive program was abandoned at one factory, the parameters examined deteriorated considerably, and a sample selected for comparison showed an abrupt drop in treatment efficiency within three years after annual screenings were discontinued.

Phage Mu d1(Ap,lac) was used to construct the hybrid crp-lac operon in Escherichia coli cells possessing crps mutation which increases sensitivity of bacterial growth to exogenous cAMP. Transcription of the structural lac genes in this hybrid operon was initiated from the promoter region of the crp gene. By measuring levels of beta-galactosidase enzyme, the maximal transcription of the crp gene was obtained in strains with cya or crp mutations causing inactivation of the CRP - cAMP complex.

Effect of hyperbaric oxygenation (oxygen pressure 3039 hPa, exposition 3 hrs) on activity of the hydroxylating system and lipid peroxidation was studied in membranes of rat liver endoplasmic reticulum. Within the first 3 hrs after hyperbaric oxygenation the rates of aniline n-hydroxylation, amidopyrine N-demethylation, NADPH oxidation and the cytochrome P-450 level were decreased, while the rates of ascorbate- and NADPH-dependent lipid peroxidation were increased. Oil emulsion of alpha-tocopherol, administered per os into rats at a dose of 50 mg/kg every 12 hrs within 2 days before hyperbaric oxygenation, prevented the posthyperoxic deterioration of xenobiotic metabolism and the intensification of lipid peroxidation in liver microsomes.

The adenylate cyclase gene of Escherichia coli has been cloned on the plasmid vector pBR325. The hybrid plasmid pTH4 obtained has a molecular weight of 6,4 megadalton and represents pBR325 plasmid with the insertion of 2,8 megadalton in the Pst1 site. The cya mutant bacteria carrying pTH4 recover their ability to utilize mannitol, lactose and other carbohydrates as carbon sources, and lose this ability again in the case of rare spontaneous excision of the DNA insert from the Pst1 site.

The genetical nature of Escherichia coli GluR mutant was studied, the mutant having been characterized by an increased basal level of enzymes of the deo-operon (thymidine phosphorylase, in particular,-up to 8-10 times, in comparison to the wild type) and of some other catabolite sensitive enzymes. The above mentioned phenotype turned out to be caused by the presence in the genome of the GluR mutant of the ptsG mutation demonstrating close linkage (98% in P1 transduction) with the crp locus. The latter was designated in this work as crp-active (crpa).

Epidemiological examination of males from 40 to 54 years of age at nine industrial enterprises revealed arterial hypertension in 19.5% of cases. As a result of intensive treatment of patients in a prophylactic cardiological department the incidence of arterial hypertension reduced, the patients learned more about their disease, the efficacy of treatment improved, and there was a decrease in losses of working time due to temporary disability of workers.
