Article Synopsis
  • Idiopathic pulmonary fibrosis (IPF) is a severe lung disease marked by excessive scarring, leading to poor respiratory function and a need for new treatment strategies, particularly involving macrophages, the key immune cells in the lungs.
  • The study aimed to explore macrophage behavior in lung slices (PCLS), which are real lung tissues cultured in a lab, to better replicate the lung environment compared to isolated cells in vitro.
  • Results showed that a polarization cocktail of specific cytokines can induce macrophage markers related to lung fibrosis in PCLS without harming the tissue, demonstrating a promising model for studying macrophage roles in IPF.

Attention-deficit/hyperactivity disorder (ADHD) is the most common childhood-onset psychiatric disorder. We investigated the effects of systemic administration of monoamine reuptake inhibitors on long-term potentiation (LTP) formation and monoamine release in the medial prefrontal cortex (mPFC) of the stroke-prone spontaneously hypertensive rat (SHRSP)/Ezo, an animal model of ADHD, and its genetic control, Wistar Kyoto (WKY)/Ezo, to elucidate the functional changes in the mPFC monoamine neural system. Methylphenidate (dopamine (DA) and noradrenaline (NA) reuptake inhibitor) and desipramine (NA reuptake inhibitor) improved LTP formation defects in the mPFC of SHRSP/Ezo, suggesting that NA or both DA and NA are required for improvement of impaired LTP.

The treatment of interstitial lung diseases, including idiopathic pulmonary fibrosis (IPF), remains challenging as current available antifibrotic agents are not effective in halting disease progression. Connective tissue growth factor (CTGF), also known as cellular communication factor 2 (CCN2), is a member of the CCN family of proteins that regulates cell signaling through cell surface receptors such as integrins, the activity of cytokines/growth factors, and the turnover of extracellular matrix (ECM) proteins. Accumulating evidence indicates that CTGF plays a crucial role in promoting lung fibrosis through multiple processes, including inducing transdifferentiation of fibroblasts to myofibroblasts, epithelial-mesenchymal transition (EMT), and cooperating with other fibrotic mediators such as TGF-β.

Approximately one-third of fibrosing interstitial lung diseases exhibit progressive pulmonary fibrosis (PPF), a clinicopathological condition distinct yet resembling idiopathic pulmonary fibrosis (IPF). PPF in ANCA-positive ILD (ANCA-ILD) is poorly documented. To clarify incidence, predictors of PPF in ANCA-ILD, and their prognostic impact, 56 patients with ANCA-ILD were followed for ≥ 1 year (April 2004 to April 2021).

Idiopathic pulmonary fibrosis (IPF) is the representative phenotype of interstitial lung disease where severe scarring develops in the lung interstitium. Although antifibrotic treatments are available and have been shown to slow the progression of IPF, improved therapeutic options are still needed. Recent data indicate that macrophages play essential pro-fibrotic roles in the pathogenesis of pulmonary fibrosis.

Nimaviridae (class Naldaviricetes) is a family of double-stranded DNA viruses infecting crustaceans, with the only officially recognized representative being white spot syndrome virus (WSSV). Chionoecetes opilio bacilliform virus (CoBV) was isolated as the causative agent of milky hemolymph disease in the snow crab Chionoecetes opilio, an economically important crustacean in the northwestern Pacific. Here, we present the complete genome sequence of CoBV and show that it is unambiguously a nimavirus.

Article Synopsis
  • - The study aims to assess the case-fatality rate (CFR) of major bleeding in patients on dual antiplatelet therapy (DAPT) after undergoing percutaneous coronary intervention (PCI) and to analyze how this rate varies based on the duration of DAPT.
  • - From a search of 2777 potential studies, 16 studies were included, revealing 823 major bleeding events with 91 being fatal among nearly 49,000 patients on DAPT, resulting in an overall CFR of 10.8%.
  • - The findings indicate that while fatal bleeding is often not reported in DAPT studies, the CFR is significant, particularly higher within the first 12 months, suggesting that shorter DAPT durations may

A 47-year-old man with symptomatic paroxysmal atrial fibrillation (AF) underwent AF ablation. Activation maps during right atrial pacing and sinus rhythm before the ablation revealed distinctive left atrial (LA) propagations with multiple LA breakthrough sites via epicardial connections. A wide area circumferential ablation was not able to achieve a right pulmonary vein (RPV) isolation and required an inner PV ablation to isolate the RPV.

  • Autotaxin (ATX) is an enzyme linked to the formation of lysophosphatidic acid (LPA), and its signaling is being explored as a treatment target for pulmonary fibrosis, particularly in fibrosing interstitial lung disease (ILD).
  • In a study of 119 patients with fibrosing ILD and 38 healthy controls, serum ATX levels were found to be significantly elevated in patients compared to controls, indicating a potential link to disease progression and prognosis.
  • The results showed that high serum ATX levels were associated with worse survival outcomes in male patients and that ATX levels decreased after acute exacerbations of idiopathic pulmonary fibrosis, suggesting it could serve as a valuable biomarker in managing fibrosing

As a candidate microRNA antifibrotic effector in skin wounds, miR-146b-5p was upregulated by basic FGF, and PDGFRα was identified as a direct target of miR-146b-5p in fibroblasts. The treatment of fibroblasts with a miR-146b-5p mimic markedly downregulated the expression of PDGFRα and collagen type I. miR-146b-5p mimic transfection in wounds markedly attenuated cutaneous fibrosis, whereas a miR-146b-5p inhibitor strongly promoted fibrosis, with increases in PDGFRα and collagen I levels.
