Article Synopsis
  • Peste des Petits Ruminants (PPR) is a disease common in Africa, and effective live attenuated vaccines (Nigeria 75/1 and Sungri/96 strains) are produced to combat it, but maintaining the cold chain for these vaccines is challenging.
  • The study assessed the thermotolerance of various stabilizer formulations for freeze-dried and reconstituted PPR vaccines by testing their viability at different temperatures over time.
  • Results indicated that the formulation with lactalbumin hydrolysate-sucrose showed the best stability, maintaining vaccine efficacy at higher temperatures, suggesting it should be the preferred stabilizer for vaccines distributed in warmer climates.

Introduction: Relatively few studies rigorously examine the factors associated with health systems strengthening and scaling of interventions at subnational government levels. We aim to examine how The Challenge Initiative (TCI) coaches subnational (state government) actors to scale proven family planning and adolescent and youth sexual and reproductive health approaches rapidly and sustainably through public health systems to respond to unmet need among the urban poor.

Methods: This mixed-methods comparative case study draws on 32 semistructured interviews with subnational government leaders and managers, nongovernmental organization leaders, and TCI Nigeria staff, triangulated with project records and government health management information system (HMIS) data.

Gnotobiotic murine models are important to understand microbiota-host interactions. Despite the role of bacteriophages as drivers for microbiome structure and function, there is no information about the structure and function of the gut virome in gnotobiotic models and the link between bacterial and bacteriophage/prophage diversity. We studied the virome of gnotobiotic murine Oligo-MM12 (12 bacterial species) and reduced Altered Schaedler Flora (ASF, three bacterial species).

The HIV-1-encoded protein Vpu forms an oligomeric ion channel/pore in membranes and interacts with host proteins to support the virus lifecycle. However, Vpu molecular mechanisms are currently not well understood. Here, we report on the Vpu oligomeric organization under membrane and aqueous conditions and provide insights into how the Vpu environment affects the oligomer formation.

Gene therapy revolves around modifying genetic makeup by inserting foreign nucleic acids into targeted cells via gene delivery methods to treat a particular disease. While the genes targeted play a key role in gene therapy, the gene delivery system used is also of utmost importance as it determines the success of gene therapy. As primary cells and stem cells are often the target cells for gene therapy in clinical trials, the delivery system would need to be robust, and viral-based entries such as lentiviral vectors work best at transporting the transgene into the cells.

Article Synopsis
  • The study explores the limitations of DNA-based microbiome analysis, particularly regarding the inability to distinguish between live and dead microorganisms.
  • A Benzonase-based approach (BDA) was tested, which effectively digests DNA from dead cells, thus providing a clearer picture of the living microbiome in skin samples.
  • The findings suggest that while BDA improves the accuracy of living microbiota assessments and reduces host DNA interference, it does not completely eliminate bias from low biomass samples.

Rabies remains a public health challenge of unknown magnitude in Liberia in spite of the goal of ensuring that no human in the country dies of rabies by 2030. The annual prevalence of Dog Bite Victims (DBVs) and true load of Annual Human Deaths (AHDs) due to rabies were not known. We investigated three selected cities of Liberia for annual prevalence of DBVs and true load of AHD due to suspected rabies, using 10-year retrospective record, 2008-2017 obtained from Buchanan, Gbarnga, and Voinjama, three socio-economically important cities in post-conflict Liberia.
