A sensitive HPLC method was developed to measure urinary N-acetylneuraminic acid (NeuAc) using a specific fluorescent labeling reagent.
The reaction for labeling NeuAc was conducted at 30°C for 30 minutes and monitored at specific wavelengths.
The method showed a detection limit of 48 fmol per injection, very low relative standard deviations for precision, and successfully analyzed diluted urine samples with concentrations ranging from 8 to 89 nmol/mg of creatinine.
