Zoonotic infections (swine-human) caused by influenza A viruses (IAVs) have been reported and linked to close contact between these species. Here, we describe eight human IAV variant infections (6 mild and 2 severe cases, including 1 death) detected in Paraná, Brazil, during 2020-2023. Genomes recovered were closely related to Brazilian swIAVs of three major lineages (1 A.

Article Synopsis
  • * Researchers utilized phylogenetic and epidemiological models to map YFV transmission patterns over different epidemic seasons and identified areas of high infection risk linked to low vaccination rates in major urban centers.
  • * By analyzing the genomic data, the study revealed three distinct YFV lineages and demonstrated the connectivity between the endemic North and the extra-Amazonian region, suggesting that genomics combined with eco-epidemiology can enhance understanding and strategies for controlling the virus.

The rapid spread of the SARS-CoV-2 Variant of Concern (VOC) Gamma in Amazonas during early 2021 fueled a second large COVID-19 epidemic wave and raised concern about the potential role of reinfections. Very few cases of reinfection associated with the VOC Gamma have been reported to date, and their potential impact on clinical, immunological, and virological parameters remains largely unexplored. Here we describe 25 cases of SARS-CoV-2 reinfection in Brazil.

Article Synopsis
  • Mosquito-borne diseases like Yellow Fever, Zika, Chikungunya, and Dengue are causing big health problems in Brazil, especially since Chikungunya started spreading in 2014.
  • In two years (2021-2022), scientists created 422 new virus genomes from 12 states to better understand how Chikungunya spreads, as there were over 312,000 reported cases.
  • They discovered that the northeast region of Brazil is the main area where the virus is spreading to other places and found that certain factors in the immune system might be changing the virus's genes in Brazil.

Recombination events have been described in the Coronaviridae family. Since the beginning of the SARS-CoV-2 pandemic, a variable degree of selection pressure has acted upon the virus, generating new strains with increased fitness in terms of viral transmission and antibody scape. Most of the SC2 variants of concern (VOC) detected so far carry a combination of key amino acid changes and indels.

Public health threats such as the current COVID-19 pandemics have required prompt action by the local, national, and international authorities. Rapid and noninvasive diagnostic methods may provide on-site detection and immediate social isolation, used as tools to rapidly control virus spreading. Accordingly, the aim of the present study was to evaluate a commercial breath analysis test (TERA.

Article Synopsis
  • Brazil has experienced high COVID-19 cases and deaths, making Latin America a major pandemic epicenter, with early sustained virus transmission and ongoing gaps in understanding.
  • Researchers analyzed over 17,000 near-complete SARS-CoV-2 genomes from across Brazil and Paraguay, finding multiple viral lineages predominantly imported from Europe before local transmission clusters emerged after November 2020.
  • Effective genomic surveillance has been crucial in tracking the real-time spread of significant variants, such as Gamma (P.1) and Zeta (P.2), and understanding cross-border virus movement into Paraguay.

Rabies is a lethal zoonosis affecting mammals worldwide. Diagnosis of rabies follows international standard protocols, primarily relying on direct immunofluorescence (DI) followed by mouse inoculation test (MIT). WHO recommends molecular biology techniques such as RT-qPCR for replacing MIT to diagnose rabies in animal samples.

Article Synopsis
  • - Brazil became a major COVID-19 epicenter in Latin America by May 2021, experiencing high transmission rates and deaths, but gaps remain in understanding how the virus spreads nationally.
  • - The study analyzes nearly complete SARS-CoV-2 genomes from Brazil and Paraguay, revealing that the initial wave was marked by multiple imported viral lineages mainly from Europe, leading to significant local transmission clusters.
  • - As the pandemic progressed without effective restrictions, Brazil saw the emergence and spread of concerning variants like Gamma and Zeta, highlighting the need for extensive genomic surveillance in South America for better pandemic management and public health strategies.

One of the most remarkable severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variants of concern (VOC) features is the significant number of mutations they acquired. However, the specific factors that drove the emergence of such variants since the second half of 2020 are not fully resolved. In this study, we describe a new SARS-CoV-2 P.

Phylogenetic analysis carried out in several Brazilian regions shows the circulation of the Asian and East-Central South African (ECSA) Chikungunya virus (CHIKV) genotypes in the country. Until now, there are no genetic studies about CHIKV strains circulating in the South region. In this study, we sequenced 5 new partial sequences of the CHIKV Envelope 1 gene from strains detected in Paraná state during the years 2016-2017.

Background: The Brazilian state of Paraná conducted a mass vaccination campaign against dengue with the tetravalent attenuated vaccine CYD-TDV. The campaign targeted thirty endemic municipalities. The objective of this study was to assess the effectiveness of CYD-TDV in preventing symptomatic virologically confirmed dengue cases according to specific age groups in five of the municipalities.

Mutations at both the receptor-binding domain (RBD) and the amino (N)-terminal domain (NTD) of the Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) Spike (S) glycoprotein can alter its antigenicity and promote immune escape. We identified that SARS-CoV-2 lineages circulating in Brazil with mutations of concern in the RBD independently acquired convergent deletions and insertions in the NTD of the S protein, which altered the NTD antigenic-supersite and other predicted epitopes at this region. Importantly, we detected the community transmission of different P.

Article Synopsis
  • Blood parasites from the Haemosporida order, like those causing malaria, are being studied for evolutionary insights, but there is limited research on wildlife parasites, particularly in the genus focused on bats.
  • This study examined 406 bats in southern Brazil using PCR to detect these parasites, finding a 1.2% positivity rate, marking the first molecular identification in common and endangered bat species in this region.
  • Phylogenetic analysis revealed that Brazilian parasite lineages are distinct and related to existing clades, prompting further research on the relationship between the parasites and their bat hosts, as well as the need for more comprehensive molecular and morphological studies.

Background: Digital PCR (dPCR) is proposed to replace real time PCR and Sanger sequencing for detection and quantification of rare mutations, frequently unnoticed in the mass of tumoral cells. Screening of endothelial growth factor receptor (EGFR) mutations is mandatory before treatment with EGFR-targeted therapy with small-molecule tyrosine kinase inhibitors, which has been approved for the treatment of advanced non-small-cell lung cancer (NSCLC).

Objective: In order to establish a cost-effective method for detection of mutations, we optimized dPCR identification of EGFR mutations in exons 18-21, and determined dPCR sensitivity, limits of detection (LoD) and quantification (LoQ).

The use of RT-LAMP (reverse transcriptase-loop mediated isothermal amplification) has been considered as a promising point-of-care method to diagnose COVID-19. In this manuscript we show that the RT-LAMP reaction has a sensitivity of only 200 RNA virus copies, with a color change from pink to yellow occurring in 100% of the 62 clinical samples tested positive by RT-qPCR. We also demonstrated that this reaction is 100% specific for SARS-CoV-2 after testing 57 clinical samples infected with dozens of different respiratory viruses and 74 individuals without any viral infection.
