To clarify the relationship between the predisposing factors and malalignment of the patello-femoral joint, a study was conducted on the basis of roentgenograms of 621 cases in which a disorder of the dérangement was suspected in at least one knee. The degree of alignment was classified into four grade groups according to the dynamic skyline view arthrograms. Each predisposing factor was expressed by patellar tendon-patella ratio, patellar facet ratio, depth index, and femoro-tibial axis angle.

Milrinone is a new bipyridine inotropic agent with direct vasodilator properties. To determine the role of the vasodilator action in mediating systemic and regional hemodynamic responses to milrinone, we administered two equipotent inotropic doses of either milrinone or dobutamine to dogs with chronic congestive right heart failure produced by tricuspid avulsion and pulmonary artery stenosis. Similar increases in cardiac output, right and left ventricular dP/dt, and left ventricular dP/dt/P were produced by milrinone and dobutamine; however, heart rate increased and mean aortic pressure decreased only with milrinone infusion.

As a possible mechanism for the absence of mitochondria-bindable hexokinase in the liver, the presence of a protease similar in action to chymotrypsin, which specifically eliminates the binding ability of the bindable hexokinase without changing its catalytic properties, was investigated in rat liver. The lysosomal fraction prepared from the liver converted the bindable hexokinase prepared from rat brain to the nonbindable form with little change in catalytic activity. The activity of such a "processing protease" was much lower in rat brain, where the bindable form is predominant.

Two histone H2B gene clones were isolated from macronuclei of Tetrahymena thermophila. Nucleotide sequences of the two clones were highly homologous within the coding region but not in the noncoding region. To determine the hemodynamic effects of isomazole, an imidazopyridine, we administered isomazole (10 and 20 micrograms/kg/min) to 10 awake dogs with right-sided congestive heart failure produced by pulmonary artery constriction and tricuspid valve avulsion. Isomazole increased cardiac output, heart rate, right ventricular and left ventricular (LV) dP/dt, LVdP/dt/P and decreased aortic pressure and total peripheral vascular resistance. Simultaneously, blood flow increased to myocardium, quadriceps muscle, brain and splanchnic beds, whereas vascular resistance decreased.

We studied the role of endogenous opiates and their interrelationships with the sympathetic nervous system in an experimental preparation of right-sided congestive heart failure (CHF) produced by surgical tricuspid avulsion and progressive pulmonary arterial constriction. Three groups of dogs with CHF and one group of sham-operated dogs were studied. One group of dogs with CHF was given normal saline as pretreatment, while the other two groups were pretreated with either propranolol alone (beta-blockade) or propranolol plus prazosin (alpha- plus beta-blockade).

Among the various biological effects of angiotensin II (AII), both pressor activity and aldosterone stimulation appear to be mediated by functionally different receptors. With this in mind, we compared pressor and aldosterone-stimulating activities of AII with those of triprolyl [(Pro)3] AII and pentasarcosyl [(Sar)5] AII. It is well known that the entry of amniotic fluid into the mother's circulation may result in the occurrence of amniotic embolism, and aspiration of meconium by the neonate may cause meconium aspiration syndrome. To study meconium-like substances detected in the urine of mother and neonate during the perinatal period, the authors carefully assayed the Urinary Meconial Index (UMI) in subjects including 100 pregnant women, 171 parturient women, 171 neonates and 68 samples of amniotic fluid. To clarify the effects of androgen on hemopoiesis in mice, we investigated changes in hemopoietic stem cells at various stages of differentiation after injection of 19-nandrolone decanoate (19-ND). 19-ND induced (a) a marked increase in the population of CFUe, especially in the spleen, and (b) a less significant increase in the number of CFUs, GM-CFC, and BFUe. The number of endogenous spleen colonies developed transiently on day 4 after irradiation (TE-CFU) increased significantly, and the initiation and induction of erythropoietic maturation was enhanced.

The nature of the hormone-binding site of detergent-solubilized angiotensin II receptor from the bovine adrenal cortex has been characterized using diaminoalkanes. Their affinity for the hormone-binding site showed a dramatic alteration depending on their chain length; at the hexamethylene stage, a maximum and competitive inhibition was observed. These results indicate that the angiotensin II receptors have a definite hydrophobic binding groove limbed with diagonally located two negative charges, and those hydrophobic and electrostatic forces may play an important role in the proper register between the receptor and hormone.

In 21 cancer patients receiving administration of FT-207 suppositories, we studied the concentration of FT-207 and 5-FU in the blood before surgery, and in the blood and tissues-tumor, normal and lymph node-sampled during surgery. These 21 cases were comprised of 12 cases of stomach cancer, 1 case of lymphosarcoma, 4 cases of rectal cancer, 2 cases of sigmoid cancer and 2 cases of breast cancer. Two hours after administration, the concentration of 5-FU reached its highest level and continued to maintain a level at which it could be expected to bring about clinical results even after four hours.
