The synthesis of peptides in solution proceeds through successive steps involving the removal of a protecting group and the formation of the peptide bond. While most methodological efforts have focused on the development of new protecting groups and coupling agents, methodologies based on minimal protecting groups have been less explored. In this research, a peptide synthesis methodology was developed using DCC and HOBt in THF-HO, avoiding the use of protecting groups, reducing reaction times, and reusing HOBt during successive couplings.
View Article and Find Full Text PDFUnlabelled: Failure of adoptive T-cell therapies in patients with cancer is linked to limited T-cell expansion and persistence, even in memory-prone 41BB-(BBz)-based chimeric antigen receptor (CAR) T cells. We show here that BBz-CAR T-cell stem/memory differentiation and persistence can be enhanced through epigenetic manipulation of the histone 3 lysine 9 trimethylation (H3K9me3) pathway. Inactivation of the H3K9 trimethyltransferase SUV39H1 enhances BBz-CAR T cell long-term persistence, protecting mice against tumor relapses and rechallenges in lung and disseminated solid tumor models up to several months after CAR T-cell infusion.

Morphology of adults and immatures of Notodontidae have been broadly investigated and many larval, pupal, and adult characters were found to be synapomorphies of subfamilies and tribes. Despite this, the current classification of Notodontidae remains unsettled as most recent classification systems are contradictory due to reliance on incomplete global sampling and, many taxa, especially in the Neotropics, are still informally classified as incertae sedis.
