PBC80 (resistant), inoculated with the anthracnose pathogen Colletotrichum gloeosporioides were examined using light, confocal laser scanning, and electron microscopy to compare the cytological differences between the compatible and incompatible interactions.
View Article and Find Full Text PDFCytoplasmic male sterility (CMS) in chili pepper is restored by one major dominant nuclear gene, restorer-of-fertility (Rf), together with some modifier genes and is also affected by temperature. As a result, male fertility was identified as having several phenotypes. That identified and used in the present study allowed partial restoration of fertility, producing plants that simultaneously produce normal and aborted pollen grains, with most grains stuck to the anther wall, even after dehiscence, resulting in low seed set per fruit.
View Article and Find Full Text PDFWe have developed a polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) marker that can distinguish male-fertile (N) and male-sterile (S) cytoplasm in onions. The PCR-RFLP marker was located in a chloroplast psbA gene amplicon. Digesting the amplicons from different cytoplasm-containing varieties with the restriction enzyme MspI revealed that N-cytoplasm plants have a functional MspI site (CCGG), whereas the S-cytoplasm plants has a substitution in that site (CTGG), and thus no MspI target.
