Background: With the rapid development of single-cell sequencing technology, histological studies are no longer limited to conventional homogenized tissues. Laser microdissection enables the accurate isolation of specific tissues or cells, and when combined with next-generation sequencing, it can reveal important biological processes at the cellular level. However, traditional laser microdissection techniques have often been complicated and time-consuming, and the quality of the RNA extracted from the collected samples has been inconsistent, limiting follow-up studies.

Cancer local recurrence increases the mortality of patients, and might be caused by field cancerization, a pre-malignant alteration of normal epithelial cells. It has been suggested that cancer-derived small extracellular vesicles (CDEs) may contribute to field cancerization, but the underlying mechanisms remain poorly understood. In this study, we aim to identify the key regulatory factors within recipient cells under the instigation of CDEs.

Article Synopsis
  • The study focuses on the C2H2 zinc finger (C2H2-ZF) gene family in citrus species, identifying 97 members in sweet orange (poly-embryonic) and 101 in pummelo (mono-embryonic).
  • It explores the phylogenetic relationships and functional differentiation within this gene family, revealing 20 differentially expressed C2H2-ZF genes linked to the embryonic types.
  • Notably, CsZFP7 is highlighted for its higher expression in poly-embryonic ovules, and its down-regulation in mini citrus leads to an increased rate of mono-embryonic seeds, suggesting its significant role in citrus embryogenesis.

Somatic embryogenesis (SE) is a key regeneration pathway in various biotechnology approaches to crop improvement, especially for economically important perennial woody crops like citrus. However, maintenance of SE capability has long been a challenge and becomes a bottleneck in biotechnology-facilitated plant improvement. In the embryogenic callus (EC) of citrus, we identified 2 csi-miR171c-targeted SCARECROW-LIKE genes CsSCL2 and CsSCL3 (CsSCL2/3), which exert positive feedback regulation on csi-miR171c expression.

Astaxanthin is a natural liposoluble ketocarotenoid with various biological activities. Hydrophobic astaxanthin with C symmetry can self-assembly form H-type aggregates and J-type aggregates in hydrated polar solvents. However, astaxanthin and its aggregates are limited by its water insolubility and chemical instability.

Background: To evaluate the performances of multiparametric MRI-based convolutional neural networks (CNNs) for the preoperative assessment of breast cancer molecular subtypes.

Methods: A total of 136 patients with 136 pathologically confirmed invasive breast cancers were randomly divided into training, validation, and testing sets in this retrospective study. The CNN models were established based on contrast-enhanced T -weighted imaging (T C), Apparent diffusion coefficient (ADC), and T -weighted imaging (T W) using the training and validation sets.

Apomixis is the phenomenon of clonal reproduction by seed. As apomixis can produce clonal progeny with exactly the same genotype as the maternal plant, it has an important application in genotype fixation and accelerating agricultural breeding strategies. The introduction of apomixis to major crops would bring many benefits to agriculture, including permanent fixation of superior genotypes and simplifying the procedures of hybrid seed production, as well as purification and rejuvenation of crops propagated vegetatively.

  • Exosomes from human umbilical cord mesenchymal stem cells (HUMSCs) may help with inflammation in rheumatoid arthritis, as shown in mouse studies.
  • Mice treated with HUMSC-derived exosomes had reduced swelling and lower levels of inflammatory markers and antibodies related to collagen.
  • The study suggests that HUMSC exosomes help restore immune balance by affecting specific immune cells and cytokines, which could offer new therapeutic approaches for rheumatoid arthritis.

Citrus nucellar poly-embryony (NPE) is a mode of sporophytic apomixis that asexual embryos formed in the seed through adventitious embryogenesis from the somatic nucellar cells. NPE allows clonal propagation of rootstocks, but it impedes citrus cross breeding. To understand the cellular processes involved in NPE initiation, we profiled the transcriptomes and DNA methylomes in laser microdissection captured citrus apomictic cells.

Grafting is an ancient technique used for plant propagation and improvement in horticultural crops for at least 1,500 years. Citrus plants, with a seed-to-seed cycle of 5-15 years, are among the fruit crops that were probably domesticated by grafting. Poncirus trifoliata, a widely used citrus rootstock, can promote early flowering, strengthen stress tolerance, and improve fruit quality via scion-rootstock interactions.

  • A study was conducted on 33 children diagnosed with congenital choledochal malformation (CM) to analyze the effects of early diagnosis and treatment between prenatally and postnatally diagnosed patients.
  • The results showed that postnatally diagnosed patients experienced more symptoms like abdominal pain and vomiting, as well as more severe histopathological findings compared to those diagnosed prenatally.
  • Despite the prenatally diagnosed group having longer surgery times and longer recovery periods, their overall hospitalization was shorter than that of the postnatally diagnosed patients.

Apomixis, is an asexual mode of seed formation resulting in genetically identical or clonal seed with a maternal genotype. Apomixis has not been reported in seed crops where its flexible application in plant breeding could accelerate delivery of new varieties. By contrast, a sporophytic form of apomixis termed nucellar or adventitious embryony is common in the Rutaceae containing Citrus crop species.

In citrus, genetic improvement via biotechnology is challenging due to insufficient understanding of molecular barriers that prevent regeneration by somatic embryogenesis (SE). Our previous study indicated that LEC genes were involved in SE in citrus, but their regulatory roles remain to be elucidated. Here, we cloned one of the LEC genes, CsFUS3, and show that it is preferentially expressed during SE and in the embryogenic callus (EC) derived from citrus varieties with strong embryogenic competence.

Because cholangiocarcinoma shows no specific clinical signs or symptoms and presents with non-specific biological and tumor markers in the early stages, MRI findings often lack typical features before this lesion becomes symptomatic and might be mistaken for other liver lesions. An evaluation of relevant radiological findings in nodular cholangiocarcinoma (≤3 cm) in stages T1N0M0 and T2N0M0 is urgently needed. In our study, we compared two groups of liver hypovascular nodules and found that a distinct margin and enhanced area/nodule size >2/3 in the delayed phase were more frequently observed in cholangiocarcinoma cases than in metastatic nodule cases in which markedly high signal intensity on T2WI was common (p < 0.

Nucellar embryony (NE) is an adventitious form of apomixis common in citrus, wherein asexual embryos initiate directly from nucellar cells surrounding the embryo sac. NE enables the fixation of desirable agronomic traits and the production of clonal offspring of virus-free rootstock, but impedes progress in hybrid breeding. In spite of the great importance of NE in citrus breeding and commercial production, little is understood about the underlying molecular mechanisms.

Wastewater irrigation can elevate metal concentrations in soils and crops and increase the metal-associated health risks via vegetable ingestion in arid and semiarid northwestern China. Here, we investigated the As, Cd, Cr, Cu, Ni, Pb, and Zn concentrations in four vegetable species from Dongdagou and Xidagou farmlands in Baiyin, Gansu, China. We evaluated the effects of irrigation type (Dongdagou: industrial wastewater; Xidagou: domestic wastewater) and cultivation mode (open field and greenhouse) on the vegetable metal concentration, metal partitioning, soil-to-plant bioconcentration factor (BCF), and the health risk index.
