Understanding the molecular cross-talk between the embryo and uterine endometrium is crucial for the improvement of IVF outcomes. The present work was undertaken to investigate the effect of pre-implantation embryo on the expression profile of immune-related genes in uterine epithelial cells (UECs) and PBMCs in buffalo. UECs were isolated from slaughterhouse-derived non-gravid uteri, cultured ex vivo and characterized, and buffalo embryos were produced in vitro from slaughterhouse-derived ovaries.

Article Synopsis
  • Buffalo spermatozoa are at risk for cryo-injury due to low antioxidant levels and high polyunsaturated fatty acids, making them susceptible to damage during freezing.
  • The study found Humanin present in buffalo spermatozoa and showed that Humanin supplementation, particularly at 5 μM, significantly improved sperm quality post-thaw, reducing lipid peroxidation and enhancing mitochondrial function and DNA integrity.
  • Results indicated that Humanin could enhance the fertility potential of buffalo sperm, as evidenced by better kinematics and lower apoptosis in treated samples compared to controls.

Background: Oxidative stress is one of the leading factors responsible for poor post-thaw semen quality because of overproduction of reactive oxygen species (ROS) over neutralizing antioxidants present in semen. Mainly two ROS generation sites are present in spermatozoa, that is, mitochondria and plasma membrane. Therefore, the idea of targeting these specific sites for minimization of ROS production with the compounds having known mechanism of actions was built up as a core for this research.
