: Previous studies have shown that intrinsic capacity changes over time and can independently predict adverse outcomes such as mortality and care dependence in older adults. However, explorations of the heterogeneity in the developmental trajectories of intrinsic capacity are limited. This study aimed to identify potential intrinsic capacity trajectory groups and the factors impacting different trajectory groups among older adults.

Purposes: Investigating the contribution of each component of the Western Aphasia Battery (WAB) to the aphasia quotient (AQ) helps better understand the mechanisms of change in the AQ. Previous studies on patients with English-speaking aphasia have shown that spontaneous speech contributes the most to the AQ. However, the same conclusion may not be applicable to Chinese-speaking patients with aphasia because of the differences between Chinese and English languages.

Background: It is difficult to identify patients with atrial fibrillation (AF) most likely to respond to ablation. While any arrhythmia patient may recur after acutely successful ablation, AF is unusual in that patients may have long-term arrhythmia freedom despite a lack of acute success. We hypothesized that acute and chronic AF ablation outcomes may reflect distinct physiology and used machine learning of multimodal data to identify their phenotypes.

Nicotinamide (NAM), a main precursor of NAD+, is essential for cellular fuel respiration, energy production, and other cellular processes. Transporters for other precursors of NAD+ such as nicotinic acid and nicotinamide mononucleotide (NMN) have been identified, but the cellular transporter of nicotinamide has not been elucidated. Here, we demonstrate that equilibrative nucleoside transporter 1 and 2 (ENT1 and 2, encoded by SLC29A1 and 2) drive cellular nicotinamide uptake and establish nicotinamide metabolism homeostasis.

Photodynamic therapy (PDT) eradicates tumor cells by the light-stimulated reactive oxygen species, which also induces lipid peroxidation (LPO) and subsequently ferroptosis, an iron-depended cell death. Ferroptosis has a tremendous therapeutic potential in cancer treatment, however, the ferroptosis efficiency is largely limited by the available iron in cells. Through hijacking the CD44-mediated iron endocytosis of hyaluronan (HA), here PDT with enhanced ferroptosis was realized by a HA@Ce6 nanogel self-assembled from HA, a photosensitizer Chlorin e6 (Ce6) and Fe as cross-linkers.

Calcium and integrin-binding protein 2 (CIB2) and CIB3 bind to transmembrane channel-like 1 (TMC1) and TMC2, the pore-forming subunits of the inner-ear mechano-electrical transduction (MET) apparatus. These interactions have been proposed to be functionally relevant across mechanosensory organs and vertebrate species. Here, we show that both CIB2 and CIB3 can form heteromeric complexes with TMC1 and TMC2 and are integral for MET function in mouse cochlea and vestibular end organs as well as in zebrafish inner ear and lateral line.

Research has shown that patients with post-stroke aphasia may encounter difficulties in producing inflectional morphemes. Interestingly, they also show distinct challenges with the inflectional morphemes of homophones, such as the plural marker and the possessive marker in English. However, previous studies have predominantly focused on morphemes in European languages.

Hibernating mammals experience severe hemodynamic changes over the torpor-arousal cycle, with oxygen consumption reaching peaks during the early stage of torpor to re-enter arousal. Melatonin (MT) can improve mitochondrial function and reduce oxidative stress and inflammation. However, the regulatory mechanisms of MT action on the vascular protective function of hibernators are still unclear.

  • Long-duration space missions to the Moon and Mars pose significant physical and mental health challenges for astronauts due to exposure to multiple stressors, particularly space radiation (SR) and sleep disturbances.
  • Ground-based studies indicate that both SR and poor sleep quality can impair cognitive performance, particularly in complex tasks like the attentional set-shifting task (ATSET).
  • This research found that fragmented sleep significantly worsened ATSET performance in rats previously exposed to galactic cosmic ray simulation (GCRsim), revealing that sleep disturbance can amplify cognitive deficits caused by SR, with variations noted between male and female rats in the type of performance decrements.

Otolith organs in the inner ear and neuromasts in the fish lateral-line harbor two populations of hair cells oriented to detect stimuli in opposing directions. The underlying mechanism is highly conserved: the transcription factor EMX2 is regionally expressed in just one hair cell population and acts through the receptor GPR156 to reverse cell orientation relative to the other population. In mouse and zebrafish, loss of Emx2 results in sensory organs that harbor only one hair cell orientation and are not innervated properly.
