To investigate the potential role of nuclear factor erythroid 2-related factor 2 (Nrf2) in licorice ethanol extract (LEE) against triptolide- (TP-) induced hepatotoxicity, HepG2 cells were exposed to LEE (30, 60, and 90 mg·L) for 12 h and then treated with TP (50 nM) for 24 h. Besides, an acute liver injury model was established in ICR mice by a single dose of TP (1.0 mg·kg, i.

Isoliquiritigenin, a flavonoid found in licorice, has been considered as an antioxidive and hepato-protective agent. Recent studies have shown that a possible mechanism for triptolide-induced hepatotoxicity is related to oxidative damage induced by reactive oxygen species. This study was done to investigate the protection effect of isoliquiritigenin against triptolide-induced hepatotoxicity and the mechanism involved.

Article Synopsis
  • Vitamin D3 is crucial for brain health, but methods to measure its metabolites in brain tissue are lacking.
  • A new analytical technique using HPLC-MS/MS was developed to measure vitamin D3 metabolites in rat serum and brain, showing high sensitivity and accuracy.
  • Results revealed that vitamin D3 levels in the brain are significantly lower with vitamin D deficiency and that serum vitamin D3 status strongly influences its availability in the brain.

Vitamin D (VD) is implicated in multiple aspects of human physiology and vitamin D receptor (VDR) polymorphisms are associated with a variety of neuropsychiatric disorders. Although VD deficiency is highly prevalent in epilepsy patients and converging evidence indicates a role for VD in the development of epilepsy, no data is available on the possible relationship between epilepsy and genetic variations of VDR. In this study, 150 controls and 82 patients with temporal lobe epilepsy (TLE) were genotyped for five common VDR polymorphisms (Cdx-2, FokI, BsmI, ApaI and TaqI) by the polymerase chain reaction-ligase detection reaction method.

Article Synopsis
  • The study investigated the relationship between temporal lobe epilepsy and the NRG1 gene polymorphisms in a group of 73 patients and 69 controls.
  • The results showed that the rs35753505 (T>C) variant was significantly associated with temporal lobe epilepsy, particularly with a higher frequency of the risk allele C in patients compared to controls.
  • The significant association was observed only in males, and no links were found with other NRG1 polymorphisms tested.

Article Synopsis
  • Licorice has detoxifying properties that can help treat drug poisoning and its side effects, though the specifics of how it works are not fully known.
  • The study found that licorice boosts the expression of certain enzymes and transporters involved in drug metabolism and detoxification, specifically phase II and phase III enzymes.
  • It suggests that licorice may activate the Nrf2 signaling pathway, which plays a key role in enhancing the body’s ability to process and eliminate drugs.

Ethnopharmacological Relevance: Licorice (Glycyrrhizae radix), the root of Glycyrrhiza uralensis Fisch. (Leguminosae), is mainly used to moderate the characteristics of toxic herbs in Traditional Chinese Medicine, which could be partly interpreted as detoxification. However, the underlying mechanism is still not fully elucidated.

Article Synopsis
  • The study investigates how vitamin D, specifically calcitriol, influences neurotransmitter systems in rat brains over a 6-week period.
  • Higher doses of calcitriol led to increased levels of GABA and enhanced the expression of related enzymes, suggesting a positive impact on GABAergic signaling.
  • Calcitriol also raised levels of glutamate and glutamine in the prefrontal cortex and influenced the expression of enzymes involved in dopamine and serotonin metabolism, indicating potential effects on mood and cognitive functions.

Depression is associated with stress-induced neural atrophy in limbic brain regions, whereas exercise has antidepressant effects as well as increasing hippocampal synaptic plasticity by strengthening neurogenesis, metabolism, and vascular function. A key mechanism mediating these broad benefits of exercise on the brain is induction of neurotrophic factors, which instruct downstream structural and functional changes. To systematically evaluate the potential neurotrophic factors that were involved in the antidepressive effects of exercise, in this study, we assessed the effects of swimming exercise on hippocampal mRNA expression of several classes of the growth factors (BDNF, GDNF, NGF, NT-3, FGF2, VEGF, and IGF-1) and peptides (VGF and NPY) in rats exposed to chronic unpredictable mild stress (CUMS).

The first HILIC-tandem mass spectrometry (MS/MS) method for determination of vindesine (VDS) in human plasma using vinorelbine as an internal standard (IS) has been developed and validated. Plasma samples clean-up consisted of solid phase extraction with a strata™-X column. The compounds were separated on a HILIC column with an isocratic mobile phase consisting of acetonitrile and 15mM ammonium acetate buffer containing 0.

  • Chronic stress and high levels of cortisol may lead to conditions like depression and heart issues, which are linked to vitamin D (VD) deficiency. This study investigates how glucocorticoids, specifically dexamethasone, impact VD in the body.
  • Rats treated with a higher dose of dexamethasone (2 mg/kg) experienced depression and a decrease in vitamin D receptor (VDR) expression and enzymes important for vitamin D metabolism in their brains, while a lower dose (0.2 mg/kg) had mixed effects on VDR expression.
  • The findings suggest that glucocorticoids like dexamethasone can disrupt the action of vitamin D in the brain and

Exposure to stressful life events is associated with the onset of major depression and increases the risk of cardiac morbidity and mortality. While recent evidence has indicated the existence of an interrelationship between local vitamin D (VD) metabolism and many aspects of human physiology including brain and heart function, much is still unknown concerning the biological link between VD signaling and stress-induced depressive behavior and cardiac dysfunction. In the present study, we observed the VD intracrine system in the hippocampus and myocardium of chronic unpredictable mild stress (CUMS) exposed rats.

Both vitamin D (VD) signaling and Nur77 are implicated in dopaminergic neurotransmission and dopamine-related neuropsychiatric disorders, such as schizophrenia and Parkinson's disease. Developmental vitamin D (DVD) deficiency rats exhibit schizophrenia-like behaviors and disturbance of dopamine system, which could be partly normalized by haloperidol treatment. By blocking dopamine D2 receptor, haloperidol induces Nur77 expression, suggesting a modulatory role of Nur77 in brain dopamine system.
