Glioblastoma (GBM) is a highly malignant brain tumor with limited treatment options. Polo-like kinase 2 (PLK2), a member of the polo-like kinase family, has been variably implicated in cancer, but its role in GBM has not been fully elucidated. We utilized RNA-seq data from multiple databases, including Gene Expression Omnibus (GEO), the Cancer Genome Atlas (TCGA), and the Chinese Glioma Genome Atlas (CGGA), and conducted experiments on human glioma cell lines to explore PLK2's expression and function.

Hemizygous genes, which are present on only one of the two homologous chromosomes of diploid organisms, have been mainly studied in the context of sex chromosomes and sex-linked genes. However, these genes can also occur on the autosomes of diploid plants due to structural variants (SVs), such as a deletion/insertion of one allele, and this phenomenon largely unexplored in plants. Here, we investigated the genomic and epigenomic landscapes of hemizygous genes across 22 genomes with varying propagation histories: eleven clonal lineages, seven outcrossed samples, and four inbred and putatively homozygous genomes.

We report a novel semi-synthetic strategy of UFM1-propargylamine by N-S acyl transfer to give UFM1R thioester and subsequent aminolysis with valine-propargylamine. The use of the valine-propargylamine molecule overcomes the inability of the valine site to efficiently undergo the N-S acyl transfer. This strategy benefits from the availability of raw materials for bulk expression, and the resulting probe shows high selectivity toward UFM1-specific proteases in buffers and cell lysates.
