Background: Pre-specified interim analyses allow for more timely evaluation of efficacy or futility, potentially accelerating decision-making on an investigational intervention. In such an analysis, the randomized, double-blind MOVe-OUT trial demonstrated superiority of molnupiravir over placebo for outpatient treatment of COVID-19 in high-risk patients. In the full analysis population, the point estimate of the treatment difference in the primary endpoint was notably lower than at the interim analysis.

Idiosyncratic drug-induced liver injury (DILI) is a leading cause of drug attrition and/or withdrawal. The formation of reactive metabolites is widely accepted as a key factor contributing to idiosyncratic DILI. Therefore, identifying reactive metabolites has become a critical focus during lead optimization, and a combination of GSH-/cyano-trapping and cytochrome P450 inactivation studies is recommended to identify compounds with the potential to generate reactive metabolites.

Glioblastoma (GBM) is one of the most lethal adult brain tumors with limited effective therapeutic options. Immunotherapy targeting B7-H3 (CD276) has shown promising efficacy in the treatment of gliomas. However, the response to this treatment varies among glioma patients due to individual differences.

Long-gauge fiber optic sensors have proven to be valuable tools for structural health monitoring, especially in reinforced concrete (RC) beam structures. While their application in this area has been well-documented, their use in RC columns remains relatively unexplored. This suggests a promising avenue for further research and development.

Land Surface Temperature (LST) is widely recognized as a sensitive indicator of climate change, and it plays a significant role in ecological research. The ERA5-Land LST dataset, developed and managed by the European Centre for Medium-Range Weather Forecasts (ECMWF), is extensively used for global or regional LST studies. However, its fine-scale application is limited by its low spatial resolution.

Background: Neuropsychiatric adverse events (NPAEs) are associated with several antiretrovirals. Doravirine (DOR), a non-nucleoside reverse transcriptase inhibitor indicated for HIV-1 treatment, does not interact significantly with known neurotransmitter receptors in vitro. First-line therapy with DOR-based regimens resulted in significantly fewer NPAEs than efavirenz/emtricitabine/tenofovir disoproxil fumarate (EFV/FTC/TDF) and similar rates to those of ritonavir-boosted darunavir (DRV/r) with 2 nucleos(t)ide reverse transcriptase inhibitors (NRTIs) through Week 96 of the phase 3 DRIVE-AHEAD and DRIVE-FORWARD studies, respectively.

The steroid constituents in the 75% ethanol extract of Cyanotis arachnoidea and their anti-HSV-1 activities in vitro were investigated in this study. The ethyl acetate fraction of the extract was isolated and purified using silica gel, ODS, Sephadex LH-20 column chromatography, and semi-preparative HPLC. Structural identification was conducted based on spectroscopic data.

Background: Dexamethasone has been widely used in treating severe COVID-19 patients due to its anti-inflammatory properties. However, its long-term impact on mortality remain unclear.

Objective: To evaluate the effect of dexamethasone on short-term (28-day) and long-term (1-year) mortality in hospitalized COVID-19 patients and to explore its efficacy across different respiratory support.

Blue phosphorescent organic light-emitting diodes (PHOLEDs) are on the brink of commercialization for decades. However, the external quantum efficiency (EQE) and operational lifetime of PHOLEDs are not yet reached industrial standards. Here, a novel tetradentate Pt(II) emitter with a spirofluorene onto the carbazole unit that minimizes the vibration modes, corresponding to the structural relaxation during the de-excitation, called the vibration suppression effect is reported.

Background: Myocardial infarction (MI) is a major disease with high morbidity and mortality worldwide. However, existing treatments are far from satisfactory, making the exploration of potent molecular targets more imperative. The E3 ubiquitin ligase RING finger protein 5 (RNF5) has been previously reported to be involved in several diseases by regulating ubiquitination-mediated protein degradation.

Anesthetic-induced brain activity study is crucial in avian cognitive-, consciousness-, and sleep-related research. However, the neurobiological mechanisms underlying the generation of brain rhythms and specific connectivity of birds during anesthesia are poorly understood. Although different kinds of anesthetics can be used to induce an anesthesia state, a comparison study of these drugs focusing on the neural pattern evolution during anesthesia is lacking.

Orthopedic surgeons treating fractures need to consider comorbidities, including chronic kidney disease (CKD), which affects millions worldwide. CKD patients are at elevated risk of fractures due to osteoporosis, especially in advanced stages. In addition, fractures in CKD patients pose challenges due to impaired bone healing and increased post-fracture complications including surgical site infection and nonunion.

Epilepsy, a chronic neuropsychiatric brain disorder characterized with recurrent seizures, is closely associated with abnormal neural communications within the brain. Despite that the phase-amplitude coupling (PAC) has been suggested to offer a new way to observe neural interactions during epilepsy, however, few studies pay attention to alterations of the epileptic functional brain network based on PAC, especially on the [Formula: see text] PAC. Therefore, we use scalp electroencephalography (EEG) data of epileptic patients and the [Formula: see text] PAC modulation index (MI) to construct functional brain networks to examine variations of neural interactions during different epileptic phases.

Article Synopsis
  • The study investigates the link between air pollution and the risk of mild cognitive impairment (MCI) in older adults in China, using data from the Hubei Memory and Aging Cohort Study.
  • Results show that higher levels of pollutants like PM2.5 and PM10 significantly increase the odds of developing MCI, particularly in older individuals.
  • The findings highlight the need for more research across different populations to further understand and address the impact of air pollution on cognitive health in aging populations.

Background: Given the lack of research on disulfidptosis, our study aimed to dissect its role in pan-cancer and explore the crosstalk between disulfidptosis and cancer immunity.

Methods: Based on TCGA, ICGC, CGGA, GSE30219, GSE31210, GSE37745, GSE50081, GSE22138, GSE41613, univariate Cox regression, LASSO regression, and multivariate Cox regression were used to construct the rough gene signature based on disulfidptosis for each type of cancer. SsGSEA and Cibersort, followed by correlation analysis, were harnessed to explore the linkage between disulfidptosis and cancer immunity.

Learning the spatial location associated with visual cues in the environment is crucial for survival. This ability is supported by a distributed interactive network. However, it is not fully understood how the most important task-related brain areas in birds, the hippocampus (Hp) and the nidopallium caudolaterale (NCL), interact in visual-spatial associative learning.

Background: Glioblastoma is one of the deadliest tumors, and limited improvement in managing glioblastoma has been achieved in the past decades. The unmethylated promoter area of 6-O-Methylguanine-DNA Methyltransferase (MGMT) is a significant biomarker for recognizing a subset of glioblastoma that is resistant to chemotherapy. Here we identified MGMT methylation can also work as a specific biomarker to classify the lipid metabolism patterns between methylated and unmethylated glioblastoma and verify the potential novel therapeutic strategy for unmethylated MGMT glioblastoma.

Due to increasingly documented health effects associated with airborne particulate matter (PM), challenges in forecasting and concern about their impact on climate change, extensive research has been conducted to improve understanding of their variability and accurately forecasting them. This study shows that atmospheric PM concentrations in Brunei-Muara district are influenced by meteorological conditions and they contribute to the warming of the Earth's atmosphere. PM predictive forecasting models based on time and meteorological parameters are successfully developed, validated and tested for prediction by multiple linear regression (MLR), random forest (RF), extreme gradient boosting (XGBoost) and artificial neural network (ANN).
