Synthetic peptides, peptides A (Arg-Leu-Tyr-Leu-Arg-Ile-Gly-Arg-Arg-NH(2)) and B (Arg-Leu-Arg-Leu-Arg-Ile-Gly-Arg-Arg-NH(2)), derived from the beetle Allomyrina dichotoma defensin, have antimicrobial activities. Immunotoxicological effect of these peptides was evaluated by cytotoxicity of mouse peritoneal macrophages. In addition, antigenicity of these peptides was studied by evaluating antibody responses in mice immunized with these peptides.

Synthetic peptides, Arg-Leu-Tyr-Leu-Arg-Ile-Gly-Arg-Arg-NH2 (peptide A) and Arg-Leu-Arg-Leu-Arg-Ile-Gly-Arg-Arg-NH2 (peptide B), derived from the beetle Allomyrina dichotoma defensin, have not only antimicrobial activities but also anti-inflammatory effects by inhibiting tumour necrosis factor-alpha(TNF-alpha) production. In the present study, we evaluated the lipopolysaccharide (LPS)-binding activities and the protective effects of these peptides on LPS-induced lethal shock in d-galactosamine (GalN)-sensitized mice. These peptides were shown to bind to erythrocytes coated with LPS and the binding activity of peptide A to LPS was significantly higher than those of peptide B and polymyxin B.

Anti-bacterial activity of two synthesized oligopeptides, RLYLRIGRR-NH2 (peptide A) and RLRLRIGRR-NH2 (peptide B), both which based on a putative active site of defensin, an anti-bacterial peptide from the beetle Allomyrina dichotoma, was examined by macroscopic and histopathologic assessment during the course of infection in mice inoculated with methicillin-resistant Staphylococcus aureus (MRSA) in vivo. Both peptides A and B decreased the mortality of mice inoculated with MRSA. Peptides A and B decreased the macroscopical and histopathological lesions by MRSA infection in mice even seven days after the challenge.

Synthetic 9-mer peptides, ALYLAIRRR-NH2 and ALYLAIRKR-NH2, designed from the amino acid sequences of active sites of insect defensins, were examined for their protective effect on Methicillin-resistant Staphylococcus aureus (MRSA) in infected mice. These peptides protected mice from lethal MRSA challenges, despite having their antibacterial activity diminished in physiological high salt conditions. These peptides suppressed the induction of tumor necrosis factor alpha gene expression by lipopolysaccharide or lipoteichoic acid in murine macrophages, contributing to protection of mice from lethal MRSA infections in vivo.

In order to determine the primary structure of vitellogenin in a protandric species, the coonstriped shrimp Pandalus hypsinotus, we previously purified four vitellin components (designated as VnA, VnB, VnC, and VnD, respectively), and chemically analyzed their partial amino acid sequences. In this study, we subsequently cloned a cDNA encoding vitellogenin in this species based on the N-terminal and internal amino acid sequences of VnA, as well as the N-terminal sequence of VnC. The open reading frame of this cDNA encoded a pro-vitellogenin in which vitellins were arranged as follows: NH2-VnA-VnB-VnC/D-COOH.

We previously reported that synthetic peptides, RLYLRIGRR-NH2 (peptide A) and RLRLRIGRR-NH2 (peptide B), derived from the beetle Allomyrina dichotoma defensin, showed antimicrobial activity against both Gram-positive and negative bacteria and suppressed lipopolysaccharide (LPS)-induced tumor necrosis factor-alpha (TNF-alpha) mRNA expression in a murine macrophage cell line. In this study, inhibitory effects of these peptides in LPS-induced mouse peritoneal macrophage activation were investigated. The supplement of peptide A to macrophages cultured with LPS resulted in a significant decrease in nitric oxide and TNF-alpha production.

Synthetic antimicrobial 9-mer peptides were designed from the amino acid sequence of an active site of insect defensin to increase the number of positively charged amino acid residues. These peptides, RLRLRIGRR-NH2, RLLLRIGRR-NH2 and RLYLRIGRR-NH2, showed strong antimicrobial activity against bacteria and fungus. These peptides showed no growth inhibition activity against murine fibroblasts or macrophages and no hemolytic activity against rabbit erythrocytes in vitro.

The dynamics of vitellogenin mRNA expression during ovarian maturation in Macrobrachium rosenbergii were examined by measuring hemolymph vitellogenin (Vg) levels and Vg mRNA expression in the hepatopancreas and ovary at differing reproductive stages in both intact and eyestalk ablated animals. Vg mRNA was quantified using real-time RT-PCR and hemolymph Vg was measured by enzyme immunoassay. In intact animals, Vg mRNA levels in the hepatopancreas and hemolymph Vg levels showed a gradual increase during the molt cycle concomitant with increasing gonadosomatic index (GSI), with Vg levels decreasing prior to ecdysis although GSI continued to increase.
