Heat-shock protein 90 (Hsp90) inhibitor downregulates c-Myc expression and upregulates the expression of tumor repressor proteins such as p53 and pRB, inhibiting the G1/S transition and causing G2/M arrest during cell cycle progression. The cycle progression is extensively controlled by the pRB/E2F signaling pathway. E2F is released from the pRB/E2F complex with the phosphorylation of pRB by cyclin-cyclin-dependent kinase (CDK) complexes.

Article Synopsis
  • Edeine B(1) is a peptide antibiotic that kills Bacillus subtilis by inducing filamentous morphology and inhibiting cell division through interaction with the FtsZ protein.
  • A temperature-sensitive mutant of FtsZ (ftsZ ts1) is more sensitive to the antibiotic compared to the parent strain, indicating that changes in the FtsZ sequence contribute to this sensitivity.
  • The variations in specific amino acid residues at positions 240, 278, 345, and 346 in the FtsZ protein among different strains suggest a link between these changes and the differing levels of susceptibility to edeine B(1), leading to cell death through disrupted FtsZ assembly and protein synthesis.

The supramolecular (HOPD(+))(DB[18]crown-6) cation, in which HOPD(+) and DB[18]crown-6 are o-aminoanilinium and dibenzo[18]crown-6, respectively, was introduced into [Ni(dmit)(2)](-) salts (dmit(2-) = 2-thioxo-1,3-dithiole-4,5-dithiolate). Conformational polymorphs were observed as tetragonal (HOPD(+))(DB[18]crown-6)[Ni(dmit)(2)](-) (1) and monoclinic (HOPD(+))(DB[18]crown-6)[Ni(dmit)(2)](-) (2). The ammonium group of HOPD(+) in salts 1 and 2 formed the N-H(+)O hydrogen bonds at the bottom and upper positions of V-shaped DB[18]crown-6, respectively, thereby producing a different supramolecular conformation.

The interaction of three photoactive organic dyes, Rhodamine B, Rhodamine 6G and a stilbazolium derivative 4'-dimethylamino-N-methyl-4-stilbazolium with synthetic sodium-saponite has been examined by UV-visible absorption spectroscopy. In all cases, bathochromic shifts and the reduction of peak absorbance for the dyes were observed in the absorption spectra at a low dye concentration (25% adsorption of the cation exchange capacity (CEC) of the clay), although the shape and the width of their absorption bands were similar to those in aqueous solution. This absorption behavior indicates that the organic dye molecules adsorbed onto the surface of the negatively charged clay particles and the adsorbed molecules were well dispersed.

Micropeptins 88-N (1) and 88-Y (2), new 3-amino-6-hydroxy-2-piperidone (Ahp)-containing cyclic depsipeptides, were isolated from Microcystis aeruginosa NIES-88. The structures were elucidated by analyses of HRFABMS, 1D and 2D NMR spectra, and chemical degradation. Micropeptins 88-N and 88-Y inhibited chymotrypsin.
