The entire nucleotide sequences (6.8 kilobase pairs) of the bphABC genes and their products involved in the initial dioxygenation and ring-meta-cleavage of biphenyls and polychlorinated biphenyls were determined. Zinc deficient bovine superoxide dismutase (Cu2E2SOD (E = empty)) was prepared and purified by high performance liquid chromatography (HPLC). Each peak was characterized as to protein, copper content and specific activity. The Cu2E2SOD peak fractionated by HPLC has a low specific activity at pH 7.

Effects of short-term administration of recombinant human erythropoietin on rat megakaryopoiesis.

Recombinant human erythropoietin (rHuEpo) was tested for its ability to stimulate rat megakaryopoiesis in vivo. Groups of Sprague-Dawley rats were injected with rHuEpo at a daily dose of 20, 80, or 200 U for 5 days. Significant thrombocytosis (a 30 to 40% increase over the control level) was found only in the rats that received 200 U/day, but some changes in the megakaryopoietic parameters were observed not only in the rats given 200 U/day, but also in those receiving 80 or 20 U/day.

We purified a protein possessing a potent ability to induce the differentiation of a murine megakaryoblastic cell line, L8057, from the supernatant of a human fibrous histiocytoma cell line, KHM-5M. The protein, a homodimer of a molecular weight of 25-kDa, has an N-terminal sequence identical with that of the beta A-chain of inhibin, indicating that it is identical to or highly homologous with activin A. Thus, it is speculated that activin A or its homologue is involved in megakaryocytic differentiation.
