Following oral administration of [14C]-gefitinib to albino and pigmented rats, radioactivity was widely and rapidly distributed, with the highest levels being found in liver, kidney, lung and gastrointestinal tract, but with only low levels penetrating the brain. Levels of radioactivity persisted in melanin-containing tissues (pigmented eye and skin). Binding to plasma proteins was high (86-94%) across the range of species examined and was 91% in human plasma.

The B chromosome polymorphism in Spanish populations of the grasshopper, Eyprepocnemis plorans (Charpentier) is ancient and widespread. Meiocytes containing B chromosomes were analyzed in our laboratory using the 3F3/2 monoclonal antibody, which binds to a kinetochore phosphoepitope whose degree of phosphorylation is sensitive to tension applied to the kinetochore. Further, the tension created by the spindle at metaphase controls a checkpoint (the metaphase checkpoint) that allows the cell to begin anaphase when all chromosomes are aligned at the metaphase plate.

Effects of drugs on the cardiovascular system are required to be assessed as part of safety pharmacology, in particular using the in vitro Human Ether-a-go-go Related Gene Product (HERG) and Purkinje fibre studies and can be used to predict safety margins prior to administration to man. Recent International Conferences on Harmonization (ICH) regulations, draft ICHS7B guidelines, indicate that levels of drug in bath solutions used should be measured if quantitative data are to be obtained for the estimation of safety margins. To accurately measure drug concentrations in bath solutions, a validated analytical method is required.

It has been hypothesized that autism results from an 'opioid peptide excess'. The aims of this study were to (1) confirm the presence of opioid peptides in the urine of children with autism and (2) determine whether dipeptidyl peptidase IV (DPPIV/CD26) is defective in children with autism. Opioid peptides were not detected in either the urine of children with autism (10 children; nine males, one female; age range 2 years 6 months to 10 years 1 month) or their siblings (10 children; seven males, three females; age range 2 years 3 months to 12 years 7 months) using liquid chromatography-ultraviolet-mass spectrometric analysis (LC-UV-MS).

Changes in patient- and therapist-rated process items, and patient-rated symptom severity assessed over a maximum of 30 sessions of psychodynamic or cognitive-behavioral psychotherapy were compared in an outpatient mental-health clinic. Patients' ratings in psychodynamic psychotherapy on two of the process items were superior to ratings by patients in cognitive-behavioral therapy, and interactions with time were discovered for an interpersonal item. Comparisons between patients who reported their initial symptom severities as most severe and those with less severe symptoms also were made, with results indicating differential effects of treatment according to perceptions of symptom severity.

Oseltamivir phosphate (Ro 64-0796/002) is a pro-drug of the anti-influenza neuraminidase inhibitor, Ro 64-0802, and as Tamiflu, has been developed for the treatment of both A and B strains of the disease. This paper describes an HPLC-MS-MS assay for both compounds in plasma and urine which fulfils all of the criteria for a good analytical method. It is sensitive with limits of quantification of 1 and 10 ng/ml for the pro-drug and active neuraminidase inhibitor, respectively.

In this article, problems associated with several methods commonly employed in research with psychiatric inpatients are discussed and the implications that these problems have for the validity of research with this population are explored, using an investigation of the relationship between moral reasoning and aggression among psychiatric inpatients as a case example. Specific issues examined include the adequacy of hospital records for diagnosing patients, the difficulty of determining when it is appropriate to approach recently admitted patients for research, and problems in the measurement of behavioral and psychological variables such as aggression and moral reasoning. Suggestions and recommendations for addressing these issues in future research are offered.

Thirty-eight directors and 334 advanced graduate students from clinical psychology programs completed a survey on ethnic minority training offered in clinical doctoral programs. Comparisons were made between directors' and students' ratings on the following variables: students' level of interest in ethnic minority training, the importance of this training, and the effectiveness of the clinical programs' minority-related education. Minority and nonminority students' responses were also compared on these variables.

Multiply charged anions of oxidized bovine insulin A-chain react with protonated quinoline exclusively by proton transfer in a Paul trap operated with helium bath gas at a pressure of 10(-3) Torr. The isomeric [C9H8N]+ ions formed from the reaction of [C4H4]+ with pyridine, on the other hand, react largely by attachment to the multiply charged anions of oxidized bovine insulin A-chain. This observation can be rationalized on the basis of competition between unimolecular decomposition versus cooling of the ion-ion collision complex.

Proton transfer from protonated pyridine to product anions derived from quadrupole ion trap collisional activation of the triply charged anion of the oligonucleotide 5'-d(AAAA)-3' and the 6- charge state of oxidized bovine insulin A-chain is shown to be a rapid and effective way to determine product charge states. The reactions are carried out in a quadrupole ion trap as part of a procedure involving three stages of mass analysis. It is demonstrated that the reactions can be driven at rates sufficiently high to convert 30-80% of the initial product anion population to second generation products in 50-200 ms.

The estimation of ranolazine, a novel piperazine derivative, and eleven of its Phase I metabolites has been undertaken by liquid chromatography-atmospheric pressure chemical ionisation mass spectrometry (LC-APCI-MS). Plasma samples, taken on day 5 of a multiple-dose study, were extracted by solid-phase extraction (SPE) and analysed, using a gradient HPLC system coupled to the APCI source of a Finnigan MAT TSQ 700 mass spectrometer. Metabolites were analysed in selected-ion monitoring (SIM) mode, using an instrument control language (ICL) procedure.

Isolated triply and doubly charged anions of the single-stranded deoxynucleotide 5'-d(AAAA)-3' were allowed to undergo ion-ion proton transfer reactions with protonated pyridine cations within a quadrupole ion trap mass spectrometer. Sufficiently high ion number densities and spatial overlap of the oppositely charged ion clouds could be achieved to yield readily measurable rates. Three general observations were made: (1) the ion-ion reaction rate constants were estimated to be 10(- (7 - 8)) cm(3) ion(-1) s(-1); (2) the ion-ion reaction rates were found to be dependent on the reactant ion number density, which could be controlled by both the reactant ion number and the pseudopotential well depth, and (3) very little fragmentation, if any, was observed, as might normally be expected with highly exothermic proton transfer reactions.

The metabolism of ranolazine (RS-43285) or (+)N-(2,6-dimethylphenyl)-4[2-hydroxy-3-(2-methoxyphenoxy)-propyl]-1- piperazine acetamide dihydrochloride was investigated in man using plasma samples obtained from four different clinical studies. The metabolite profiles following single and multiple doses of 342 mg instant release (IR) ranolazine, following multiple doses of 1000 mg sustained release (SR) ranolazine and following dosing with both ranolazine (IR) and a potentially co-administered drug, diltiazem, were compared. Metabolism of ranolazine in man was shown by LC/MS analysis to be extensive with up to seven primary routes of metabolism identified.

Presented here is an exploration of the motivations involved in the development and application of managed mental health care to the private practice of outpatient psychotherapy. The interaction of management policy and psychotherapy is conceptualized in a dynamic model designed to provide insights into effective care policies. The model is described first, then the development of managed care, which appears defensive and is becoming symptomatic to the point of needing significant change.
