The serum neutralization (SN) test has been regarded as the "gold standard" for seroconversion following foot-and-mouth disease virus (FMDV) vaccination, although a high-level biosafety laboratory is necessary. ELISA is one alternative, and its format is constantly being improved. However, recent studies indicated that the antibodies of vaccinated farm animals are mainly against site 2 rather than site 1. In Taiwan, blanket vaccination had been implemented for more than fifteen years, in which the porcinophilic isolate O/Penghu/2012 showed significant amino acid alterations in site 2 compared to the early isolate O/TW/97.
