An emerging concept in cell signalling is the natural role of reactive oxygen species such as hydrogen peroxide (H2O2) as beneficial messengers in redox signalling pathways. The nature of H2O2 signalling is confounded, however, by difficulties in tracking it in living systems, both spatially and temporally, at low concentrations. Here, we develop an array of fluorescent single-walled carbon nanotubes that can selectively record, in real time, the discrete, stochastic quenching events that occur as H2O2 molecules are emitted from individual human epidermal carcinoma cells stimulated by epidermal growth factor.

A major challenge in the synthesis of nanotube or nanowire sensors is to impart selective analyte binding through means other than covalent linkages, which compromise electronic and optical properties. We synthesized a 3,4-diaminophenyl-functionalized dextran (DAP-dex) wrapping for single-walled carbon nanotubes (SWNTs) that imparts rapid and selective fluorescence detection of nitric oxide (NO), a messenger for biological signalling. The near-infrared (nIR) fluorescence of SWNT(DAP-dex) is immediately and directly bleached by NO, but not by other reactive nitrogen and oxygen species.

Study Objective: Our aim was to determine the diagnostic significance of the association between the abdominal skin bruise from a seat belt and the presence of intraabdominal injury.

Methods: This was an observational analysis of prospectively collected data on 45 patients who presented with an abdominal seat belt sign to a level 1 trauma center following a motor vehicle accident between July 2004 and December 2007. The 1D quantum confinement of photogenerated excitons in single-walled carbon nanotubes (SWNT) can amplify the detection of molecular adsorption to where single-molecule discrimination is realizable, even from within living cells and tissues. Toward this aim, we present a type 1 collagen film, similar to those used as 3D cell scaffolds for tissue engineering, containing embedded SWNT capable of reporting single-molecule adsorption of quenching molecules. We utilize hidden Markov modeling to link single-molecule adsorption events to rate constants for H2O2, H+, and Fe(CN)6(3-).

By reaction of the dichloride rac-(ebthi)HfCl(2) [ebthi = 1,2-ethylene-1,1'-bis(eta(5)-tetrahydroindenyl)] with lithium in the presence of bis(trimethylsilyl)acetylene, the hafnacyclopropene rac-(ebthi)Hf(eta(2)-Me(3)SiC(2)SiMe(3)) (1-Hf) was obtained. The reaction of the blue-green complex 1-Hf with an excess of ethylene at room temperature leads by insertion of the olefin to the yellow-green hafnacyclopentene 2-Hf which is only stable in solution and eliminates the alkyne at 100 degrees C under ethylene to form the corresponding yellow hafnacyclopentane 3-Hf, which was characterized by X-ray crystal structure analysis. The reaction of 1-Hf to give stepwise via 2-Hf the complex 3-Hf was investigated in detail and compared to the formation and stability of the corresponding zirconacyclopropene 1-Zr, zirconacyclopentene 2-Zr, and zirconacyclopentane 3-Zr.

The cellular uptake and expulsion rates of length-fractionated single-walled carbon nanotubes (SWNT) from 130 to 660 nm in NIH-3T3 cells were measured via single particle tracking of their intrinsic photoluminescence. We develop a quantitative model to correlate endocytosis rate with nanoparticle geometry that accurately describes this data set and also literature results for Au nanoparticles. The model asserts that nanoparticles cluster on the cell membrane to form a size sufficient to generate a large enough enthalpic contribution via receptor ligand interactions to overcome the elastic energy and entropic barriers associated with vesicle formation.

Nanoscale sensing elements offer promise for single-molecule analyte detection in physically or biologically constrained environments. Single-walled carbon nanotubes have several advantages when used as optical sensors, such as photostable near-infrared emission for prolonged detection through biological media and single-molecule sensitivity. Molecular adsorption can be transduced into an optical signal by perturbing the electronic structure of the nanotubes.

Background: Programmatic data from New York City syringe exchange programs suggest that many clients visit the programs infrequently and take few syringes per transaction, while separate survey data from individuals using these programs indicate that frequent injecting - at least daily - is common. Together, these data suggest a possible "syringe gap" between the number of injections performed by users and the number of syringes they are receiving from programs for those injections.

Methods: We surveyed a convenience sample of 478 injecting drug users in New York City at syringe exchange programs to determine whether program syringe coverage was adequate to support safer injecting practices in this group.

We developed a new method to analyze animal feed and feed ingredients for melamine and cyanuric acid. The method is capable of extracting and detecting both melamine and cyanuric acid in a single procedure, whether present as free compounds or bound together as the melamine:cyanurate complex. A novel chromatographic system based on zwitterionic hydrophilic interaction chromatography (ZIC-HILIC) columns enables separation and detection of both compounds in one run.

Three experiments re-examined Baylis and Driver's (1993) strong evidence for object-based selection, that making relative apex location judgments is harder between two objects than within a single object, with object (figure-ground) segmentation determined solely by color-based perceptual set. Using variations of the Baylis and Driver paradigm, the experiments replicated a two-object cost. However, they also showed a large part of the two-object cost to be attributable to space-based factors, though there remained an irreducible cost consistent with 'true' object-based selection.

Various trinuclear rhodium complexes of the type [Rh(3)(PP)(3)(mu(3)-OH)(x)(mu(3)-OMe)(2-x)]BF(4) (where PP = Me-DuPhos, dipamp, dppp, dppe; different ligands and mu-bridging anions) are presented, which are formed upon addition of bases such as NEt(3) to solvate complexes [Rh(PP)(solvent)(2)]BF(4). They were extensively characterized by X-ray diffraction and NMR spectroscopy ((103)Rh, (31)P, (13)C, (1)H). Their in situ formation resulting from basic additives (NEt(3)) or basic prochiral olefins (without addition of another base) can cause deactivation of the asymmetric hydrogenation.

Objective: To determine whether renal crystals can be experimentally induced in animals fed melamine or the related triazine compound cyanuric acid, separately or in combination, and to compare experimentally induced crystals with those from a cat with triazine-related renal failure.

Animals: 75 fish (21 tilapia, 24 rainbow trout, 15 channel catfish, and 15 Atlantic salmon), 4 pigs, and 1 cat that was euthanatized because of renal failure.

Procedures: Fish and pigs were fed a target dosage of melamine (400 mg/kg), cyanuric acid (400 mg/kg), or melamine and cyanuric acid (400 mg of each compound/kg) daily for 3 days and were euthanatized 1, 3, 6, 10, or 14 days after administration ceased.
