Proteins can be rapidly prototyped with cell-free expression (CFE) but in most cases there is a lack of probes or assays to measure their function directly in the cell lysate, thereby limiting the throughput of these screens. Increased throughput is needed to build standardized, sequence to function data sets to feed machine learning guided protein optimization. Herein, we describe the use of fluorescent single-walled carbon nanotubes (SWCNT) as effective probes for measuring protease activity directly in cell-free lysate.

Background: Malaria parasites have gradually developed resistance to commonly used antimalarial drugs. For decades, chloroquine was the most widely used drug to eradicate malaria. However, with the spread of chloroquine resistance, many countries have adopted combination therapies that utilize two drugs acting synergistically instead of monotherapy.

In this work, Oxford Nanopore sequencing is tested as an accessible method for quantifying heterogeneity of amplified DNA. This method enables rapid quantification of deletions, insertions, and substitutions, the probability of each mutation error, and their locations in the replicated sequences. Amplification techniques tested were conventional polymerase chain reaction (PCR) with varying levels of polymerase fidelity (OneTaq, Phusion, and Q5) as well as rolling circle amplification (RCA) with Phi29 polymerase.

Article Synopsis
  • Kinesiophobia, or the fear of movement, negatively affects daily activities, often resulting from past traumatic experiences or observations of pain in others.* -
  • A study in 2023 assessed the Persian version of the Tampa Scale for Kinesiophobia among 400 Iranian post-surgery patients, focusing on its psychometric validity through various analyses.* -
  • The results confirmed strong reliability and validity of the scale, including two main factors that accounted for over 60% of variance, making it a useful tool for understanding and managing kinesiophobia in Iran.*

Background: The trace element of zinc (Zn) has shown great effectiveness in control of leishmaniasis infection. Hence, the present study conducted a systematic review of in vitro and in vivo studies evaluating the zinc effect in the treatment or prevention of leishmaniasis.

Materials And Methods: A systematic literature search was performed of all articles published in PubMed, SciELO, ScienceDirect, Scopus, Google Scholar, and Web of Science databases (1997-2023).

Background: Cutaneous leishmaniasis (CL) infection is caused by the parasite and affects 1.5 to 2 million people worldwide each year. Although research into vaccines and antiparasitic drugs has been somewhat successful, their adverse effects include high toxicity, prolonged regeneration, and scarring.

Background: With the recent surge in the utilization of electronic health records for cognitive decline, the research community has turned its attention to conducting fine-grained analyses of dementia onset using advanced techniques. Previous works have mostly focused on machine learning-based prediction of dementia, lacking the analysis of dementia progression and its associations with risk factors over time. The black box nature of machine learning models has also raised concerns regarding their uncertainty and safety in decision making, particularly in sensitive domains like healthcare.

Article Synopsis
  • Leishmaniasis is caused by protozoan parasites from the genus Leishmania, and this study investigates the anti-leishmanial effects of a plant extract (Rech.f) on these parasites.
  • The plant material was collected in Iran and extracted with various solvents for testing on different forms of Leishmania and macrophage cells, using assays to measure cell viability and apoptosis.
  • Results showed significant anti-leishmanial effects of the methanolic fraction over time, with treated cells exhibiting morphological changes and reduced parasite numbers, suggesting the plant extract has promising therapeutic potential.

Article Synopsis
  • Current arrhythmia treatments like ion channel blockers and defibrillators have drawbacks, prompting the need for new strategies to manage heart rhythm issues.
  • A promising approach involves monitoring and adjusting specific ion levels, particularly zinc, to prevent or treat arrhythmias due to its significant impact on heart electrical activity.
  • Zinc plays a crucial role by interacting with various ion channels and receptors, influencing oxidative stress, inflammation, and even circadian rhythms, highlighting the necessity of maintaining proper zinc levels in the body for heart health.

Arsenic is a metalloid found in the environment that causes toxic effects in different organs, mainly the liver. This study aimed to investigate the protective effects of epicatechin (EC), a natural flavonol, on glucose intolerance (GI) and liver toxicity caused by sodium arsenite (SA) in mice. Our findings showed that SA exposure led to the development of GI.

Ecofriendly hydrogels were prepared using chitosan (CH, 285 kDa) and two fractions of low molecular weight microbial poly-γ-glutamic acid (γ-PGA) (R1 and R2 of 59 kDa and 20 kDa, respectively). The hydrogels were synthesized through sustainable physical blending, employing three CH/γ-PGA mass ratios (1/9, 2/8, and 3/7), resulting in the formation of physically crosslinked materials. The six resulting CH/R1 and CH/R2 hydrogels were physico-chemically characterized and the ones with the highest yields (CH/R1 and CH/R2 ratio of 3/7), analyzed for rheological and morphological properties, showed to act as bio-glues on wood and aluminum compared to commercial vinyl- (V1) and acetovinyl (V2) glues.

As various contextual and individual difference factors determine how and when mindsets may influence learning outcomes, burgeoning L2 research has recently addressed the role of growth language mindset (GLM) in different learning outcomes such as L2 Willingness to Communicate (WTC). Since little is known about the underlying mechanism through which GLM may contribute to WTC, a highly desirable goal of L2 education and an important criterion for assessing its efficiency and success, the present study addresses this gap by investigating the possible mediating and moderating roles of linguistic risk taking and L2 learning experience, respectively. The participants were 392 Iranian L2 students chosen by multi-stage cluster sampling.
