Cytotoxicity of 3-O-(beta-D-glucopyranosyl) etioline, a steroidal alkaloid from Solanum diphyllum L.

In continuation of our interest in phytochemical screening of the Egyptian flora for potential drugs, the reinvestigation of the methanolic extract of the roots of Solanum diphyllum, which grows naturally in the south of Egypt and is recorded as new to the Egyptian flora, afforded an interesting, highly cytotoxic compound, named 3-O-(beta-D-glucopyranosyl) etioline [(25S)-22,26-epimino-3beta-(beta-D-glucopyranosyloxy) cholesta-5,22(N)-dien-16alpha-ol]. The chemical structure of this compound was determined by comprehensive NMR studies, including DEPT, COSY, HMQC, and MS. The compound exhibited high cytotoxic effects against the cervical cancer cell line, Hela cells, with an IC50 value of 150 microg/mL.

Bioactive jatrophane diterpenes from Euphorbia guyoniana.

Chromatographic investigation of the methylenechloride/methanol extract of the aerial parts of Euphorbia guyoniana afforded two jatrophane diterpenes, designated guyonianins E and F, in addition to a known jatrophane diterpene. The structures of the compounds were determined by comprehensive NMR analyses, including DEPT, COSY, HMQC, HMBC, NOESY and HRMS. These compounds exhibited cytotoxicity against human embryonic kidney 293 (HEK293) cells with IC(50) values of 35-100 microM.

Biological activity of a phloroglucinol glucoside derivative from Conyza aegyptiaca.

The phloroglucinol glucoside derivative [2,4-dihydroxy-6-(beta-D-glucopyranosyloxy)phenyl]-butan-1-one (1), roseoside (2), and kaempferol-3-O-beta-D-glucopyranoside (3) were isolated from the aerial parts of Conyza aegyptiaca (L.). To the best of our knowledge, this is the first isolation of compounds 1-3 from C.

Rare prenylated flavonoids from Tephrosia purpurea.

Chemical investigations of aerial parts of Tephrosia purpurea yielded the rare prenylated flavonoids, tephropurpulin A (1) and isoglabratephrin (2), in addition to a previously identified flavonoid, glabratephrin (3). Structures were established by 1D and 2D NMR spectroscopy, as well as by HR-MS analysis; for compounds 2 and 3, structures were confirmed by X-ray analysis.

Turrealabdane, turreanone and an antisalmonellal agent from Turraeanthus africanus.

Phytochemical investigation of the seeds of Turraeanthus africanus (Meliaceae), a Cameroonian plant species traditionally used in the treatment of typhoid fever, afforded eight compounds, including two labdanes, a C-arabinoside derivative, a sesquiterpene, and several triterpenes, two of which are new: 15',16'-dihydroxy-15(12'),15'(16')-diolidebislabd-8(17),8'(17'),12-trien-16-al ( 1), trivially named turrealabdane and a C-arabinoside derivative ( 2), trivially named turreanone. The other compounds are 12,15-epoxylabda-8(17),12,14-trien-16-acetate ( 3), (+)-eudesmanol- O-L-arabinoside, cyclolaudenol, stigmasterol, sitosterol glucoside and lupeol. Acetylation and oxidation of turrealabdane yielded 15',16'-diacetoxyturrealabdane and 15,12'-dioxoturrealabdane-15',16'-dial, respectively.

Results of treatment of idiopathic clubfoot in older infants using the Ponseti method: a preliminary report.

The Ponseti method has become a well-established technique for the treatment of clubfoot presenting in the neonatal period. A few reports have discussed the result of this method in older age group. The purpose of this study is to present the results and clinical experience of using the Ponseti method in the treatment of idiopathic congenital talipes equinovarus in infants presented between 4 and 13 months of age with a history of failed manipulations.

Protein profile and morphometry of cultured human Blastocystis hominis from children with gastroenteritis and healthy ones.

A total of 180 children of age group 5-12 years old in both sexes, of whom 90 were symptomatic and negative for other parasites, rotavirus or pathogenic bacteria. Another 90 children were asymptomatic, but with B. hominis in stools.

Ketoisophorone transformation by Marchantia polymorpha and Nicotiana tabacum cultured cells.

Stereospecific olefin (C=C) and carbonyl (C=O) reduction of the readily available prochiral compound ketoisophorone (2,2,6-trimethyl-2-cyclohexene-1,4-dione) (1) by Marchantia polymorpha and Nicotiana tabacum cell suspension cultures produce the chiral products (6R)-levodione (2), (4R,5S)-4-hydroxy-3,3,5-trimethylcyclohexanone (3), and (4R,6R)-actinol (4) as well as the minor components (4R)-hydroxyisophorone (5) and (4S)-phorenol (6).

Cyclooxygenase (COX)-1 and -2 inhibitory labdane diterpenes from Crassocephalum mannii.

Two new labdane diterpenes, 8alpha,19-dihydroxylabd-13 E-en-15-oic acid (1) and 13,14,15,16-tetranorlabdane-8alpha,12,14-triol (2), as well as an acetylated derivative, 8alpha-O-beta-D-glucopyranosyllabd-13 E-ene-15,19-diol-8alpha-2',3',4',6'-hexaacetate (3a), were isolated from the aerial parts of Crassocephalum mannii. The structures of 1, 2, and 3a were elucidated by spectroscopic data analysis. Selective inhibitory activity for 1 and 2 and their acetate derivatives, 1a and 2a, against cyclooxygenases (COX-1 and COX-2) was detected.

Transcriptome analysis of agmatine and putrescine catabolism in Pseudomonas aeruginosa PAO1.

Polyamines (putrescine, spermidine, and spermine) are major organic polycations essential for a wide spectrum of cellular processes. The cells require mechanisms to maintain homeostasis of intracellular polyamines to prevent otherwise severe adverse effects. We performed a detailed transcriptome profile analysis of Pseudomonas aeruginosa in response to agmatine and putrescine with an emphasis in polyamine catabolism.

The diagnostic efficacy of urinary TGF-beta1 and VEGF in bladder cancer: comparison with voided urine cytology.

The purpose of this study was to evaluate the diagnostic efficacy of urinary transforming growth factor-beta1 (TGF-beta1) and vascular endothelial growth factor (VEGF) in comparison with voided urine cytology in the detection of bladder cancer. This study included 120 patients with bladder cancer, 54 patients with benign urological disorders and 55 healthy volunteers. Urine supernatant was used for estimation of TGF-beta1 and VEGF by ELISA.

Echinococcosis granulosus/hydatidosis an endemic zoonotic disease in Egypt.

The hydatidosis patients were collected from Mansoura University's Hospitals and Dakahlia Public Health Hospitals. The patients were divided into three groups: GI: 74 patients with surgically confirmed cystic echinococcosis (CE), GII: 45 patients with other parasitic infections, GIII: 30 healthy parasite-free individuals. All were subjected to questionnaire and full clinical examination, imaging investigation, routine laboratory examination, and serum samples for antibody against CE by the specific ELISA.

Echinococcosis granulosus in stray dogs and Echino-IHAT in the hunters in Cairo, Egypt.

A total of fifty stray dogs of both sex and of different ages were examined for natural infection with Echinococcus granulosus after post-mortem. Three main intestinal helminthes were recovered. These were in descending order of abundance Dipylidium caninum, Toxocara canis and E.

Characteristics of patients with congenital clasped thumb: a prospective study of 40 patients with the results of treatment.

Purpose: Congenital clasped thumb is a deformity that is associated with heterogeneous congenital anomalies and it has been addressed in many congenital syndromes. The aim of this study was to diagnose and evaluate cases of clasped thumb as regards the associated congenital anomalies and syndromes, and evaluation of the results of treatment of such cases.

Methods: A prospective study on 40 patients with 73 clasped thumbs was done.

Minimally invasive video-assisted thyroidectomy for small follicular thyroid nodules.

Minimal access surgery in the thyroid compartment has evolved considerably over the past 10 years and now takes many forms. This study examined the feasibility and reliability of minimally invasive thyroid surgery for the management of small benign thyroid lesions. A total of 68 patients with small thyroid nodules admitted to the Oncology Center of Mansoura University, Egypt, were enrolled in this prospective randomized trial.

Acylated pregnane glycosides from Caralluma russeliana.

The chloroform extract of the aerial parts of Caralluma russeliana yielded four acylated pregnane glycosides, namely russeliosides E-H, three were found now. The structures of the compounds were elucidated using MS, 1H NMR, 13C NMR, 1H-1H COSY, HMQC, NOESY and HMBC experiments.

Ferulsinaic acid, a sesquiterpene coumarin with a rare carbon skeleton from Ferula species.

Fractionation of methylene chloride extracts of the resin of Ferula vesceritensis and F. sinaica afforded three sesquiterpene coumarins and a glucose derivative. One of them was a sesquiterpene with a rare carbon skeleton.

The impact of polymerase chain reaction assays for the detection of hepatitis C virus infection in a hemodialysis unit.

Hepatitis C virus (HCV) infection is most often diagnosed by detection of antibodies against the virus (HCV Ab). However, it has been reported that some HCV Ab negative patients test positive for HCV-RNA. Over a study period of 30 months, all patients on hemodialysis at the Al Hada Armed Forces Hospital in Taif, Saudi Arabia were tested monthly for HCV Ab and twice per year for HCV-RNA.

Preoperative isolated limb infusion of Doxorubicin and external irradiation for limb-threatening soft tissue sarcomas.

Background: At present, limb-sparing surgery is the most appropriate and acceptable treatment option for soft tissue sarcomas of the extremities. To increase the number of limb-sparing resections in the treatment of locally advanced soft tissue sarcomas of the extremities, preoperative radiotherapy and/or chemotherapy are often used. Isolated limb perfusion of cytostatic agents is an effective alternative option but technically complex.

Constituents of Chrysothamnus viscidiflorus.

A phytochemical investigation of the aerial parts of Chrysothamnus viscidiflorus var. viscidiflorus afforded three new [chrysothol (1), 2 and 4] and seven known compounds, including five sesquiterpenes, two cinnamic acid derivatives, two ketoalcohol derivatives and one coumarin glucoside. The structures of two previously reported compounds, 1b and 1c, were revised on the basis of chemical reaction.

Neurophysiologic changes in live related kidney transplant children and adolescents.

Uremic neuropathy is one of the most debilitating symptoms associated with end stage renal disease. In adults the only potential cure for uremic neuropathy is renal transplantation. No studies have investigated the neurophysiologic abnormalities among pediatric renal transplant recipients.

Neoadjuvant chemotherapy versus primary surgery in advanced ovarian carcinoma.

Background: Patients with advanced ovarian cancer should be treated by radical debulking surgery aiming at complete tumor resection. Unfortunately about 70% of the patients present with advanced disease, when optimal debulking can not be obtained, and therefore these patients gain little benefit from surgery. Neoadjuvant chemotherapy (NACT) has been proposed as a novel therapeutic approach in such cases.

Diagnostic potentials of copro-antigen detection based ELISA, compared to microscopy in intestinal amoebiasis.

Correction of relapsed or neglected clubfoot using a simple Ilizarov frame.

We present the results of using a simple Ilizarov fixator frame in treatment of 66 feet in 52 patients (mean age 8.5 years) of 58 relapsed and eight neglected clubfeet with grade III or IV severity with a mean follow-up of 40 (26-58) months. Our frame, in spite of being simple and easy for surgeons and patients to handle, achieved satisfactory correction comparable to the literature.

Flavonoids and terpenoids from the resinous exudates of Madia species (Asteraceae, Helenieae).

The resinous material accumulated on aerial parts of Madia species is shown to consist mainly of diterpenes, containing a series of flavonoid aglycones. A6- and/or 8-O-substitution is characteristic for many of these flavonoids. Three known rare diterpenes were found and the structure elucidation of a diterpene with a new carbon skeleton, named madiaol, is reported.
