Functional avidity is supposed to critically shape the quality of immune responses, thereby influencing host protection against infectious agents including SARS-CoV-2. Here we show that after human SARS-CoV-2 vaccination, a large portion of high-avidity spike-specific CD4+ T cells lost CD3 expression after in vitro activation. The CD3- subset was enriched for cytokine-positive cells, including elevated per-cell expression levels, and showed increased polyfunctionality.

Here, we reveal a new mechanism used by glioma cells to modulate TMZ sensitivity via regulation of SORBS2 and DDR1 genes by super-enhancer RNA LINC02454. We report that LINC02454 activity increases glioma cell TMZ sensitivity by maintaining long-range chromatin interactions between SORBS2 and the LINC02454 enhancer.

Purpose: To examine whether body fat rate (BF%) is associated with cardiorespiratory fitness (CRF) and whether cardiorespiratory fitness (CRF) mediates the association between BF% and Executive function (EF) in young adults.
Methods: In this cross-sectional study, 226 college students were recruited from an university. Flanker, 2-back, and odder and shifting tasks were used to assess EF.

Here, we first develop uCoTarget, utilizing a split-pool barcoding strategy for realizing ultrahigh-throughput single-cell joint profiling of multiple epigenetic proteins. Through extensive optimization for sensitivity and multimodality resolution, we demonstrate that uCoTarget enables simultaneous detection of five histone modifications (H3K27ac, H3K4me3, H3K4me1, H3K36me3, and H3K27me3) in 19,860 single cells.

A "turn-on" aptasensor for label-free and cell-free EpCAM detection was constructed by employing magnetic α-FeO/FeO@Au nanocomposites as a matrix for signal amplification and double-stranded complex (SH-DNA/Apt probes) immobilization through Au-S binding. α-FeO/FeO@Au could be efficiently assembled into uniform and stable self-assembly films via magnetic-induced self-assembly technique on a magnetic glassy carbon electrode (MGCE). Multi-antigen stimulated cell therapy-I (MASCT-I) technology is a sequential immune cell therapy for cancer, which composes of multiple antigen-loaded dendritic cell (DC) vaccines followed by the adoptive transfer of anti-tumor effector T-cells.
Methods: In this phase 1 study, we assessed MASCT-I plus camrelizumab (an ICI against PD-1) and apatinib (a highly selective TKI targeting VEGFR2) in patients with unresectable recurrent or metastatic bone and soft-tissue sarcoma after at least one line of prior systemic therapy.

Current Background-oriented schlieren tomography (BOST) methods rely primarily on iterative algorithms for reconstruction. Before reconstruction, a weight projection matrix was generated by performing 3D ray tracing using the projection relationship between the cameras, depending on the camera calibration parameters and large weight projection matrix which introduce artifacts and greatly reduce computational efficiency in the reconstruction. Considering that CT reconstruction uses spatial projection sequences from multiple directions, this study draws inspiration from the Recurrent Neural network(RNN) and utilizes spatial correlation between adjacent projection data to propose a background-oriented schlieren reconstruction method based on a gated recurrent unit (GRU) neural network.
