Enteroviruses can infect various human organs, causing diseases such as meningitis, the common cold, hand-foot-and-mouth disease, myocarditis, pancreatitis, hepatitis, poliomyelitis, sepsis, and type 1 diabetes. Currently, there are no approved treatments for enterovirus infections. In this study, we identified a synergistic combination of orally available, safe-in-man pleconaril, AG7404, and mindeudesivir, that at non-toxic concentrations effectively inhibited enterovirus replication in human cell and organoid cultures.

Objective: To generate a list of candidate items potentially useful for discriminating individuals with Early-stage Symptomatic Knee Osteoarthritis (EsSKOA) from those with other conditions and from established osteoarthritis (OA), and to reduce this list based on expert consensus.

Design: We conducted a three-round online international modified Delphi exercise with OA clinicians and researchers ("OA experts"). In Round 1, participants reviewed 84 candidate items and nominated additional item(s) potentially useful for EsSKOA classification; those nominated by ≥3 participants were added.

Article Synopsis
  • The study aimed to see if changes in MRI-defined bone marrow lesions (BMLs) and inflammatory markers are linked to cartilage loss in knees with osteoarthritis over 24 months.
  • It analyzed data from 629 participants, finding that knees with no BMLs showed significantly less cartilage loss, while those with increasing BML size experienced more cartilage loss.
  • The results suggest that preventing the growth of BMLs and worsening of Hoffa-synovitis could help reduce cartilage deterioration in osteoarthritis patients.

Article Synopsis
  • The study looked at people with and without hand osteoarthritis to find out who has marginal erosions (MEs), which are small damage spots in the bones.
  • It discovered that MEs are more common in older men who smoke and usually happen in joints that don’t have a lot of arthritis.
  • Lastly, MEs could help predict how the condition might get worse over time, since they are linked to more changes in bone damage compared to joints without MEs.

Objective: Hand osteoarthritis (OA) pain is characterized as heterogeneous and multifactorial. Differences in pain may be explained by underlying phenotypes, which have not been previously explored DESIGN: Latent class analysis determined classes of participants with hand OA from the Nor-Hand study baseline examination (2016-17) based on a biopsychosocial framework. Outcomes were hand and overall bodily pain intensity (Numeric Rating Scale, 0-10) at baseline and follow-up (2019-21), The relations of the classes to pain outcomes at baseline, follow-up, and change over time were analysed in separate models by linear regression, using the overall healthiest class as reference.

Article Synopsis
  • The study introduces a deep learning framework using attention mechanisms to predict the progression of patellofemoral osteoarthritis (PFOA) over seven years, based on knee X-ray imaging.
  • It involved 1,832 subjects and utilized an automated tool for analyzing knee images, comparing the deep learning model's performance against traditional risk factors like age and BMI.
  • Results indicated that the deep learning model with attention outperformed other models, achieving an AUC of 0.856, and combining imaging and clinical data in an ensemble model provided slightly better predictions with an AUC of 0.865.

Persons with schizophrenia often show executive dysfunction assessed with both subjective (self-report) and objective (neuropsychological tests) measures. In a recent randomized controlled trial (RCT), subjective executive functioning in everyday life was improved following Goal Management Training (GMT). The aim of the current study is to investigate the potential of GMT to improve secondary well-being outcomes from that RCT, including self-esteem, self-efficacy, and quality of life in persons with schizophrenia spectrum disorders.

Article Synopsis
  • The study aims to analyze cytokine levels in the tear fluid of patients undergoing multiple intravitreal injections (IVI) of anti-VEGF for treating neovascular age-related macular degeneration (nAMD).
  • Researchers measured the levels of six cytokines in the tears of treated and untreated eyes of patients, using a specific assay technique, at least four weeks after receiving an injection.
  • Results showed that levels of two pro-inflammatory cytokines, IFN-γ and TNF, were lower in the injected eyes compared to the untreated eyes, supporting the notion that serial IVI treatment correlates with reduced inflammation and improved tear film health.

Article Synopsis
  • Erosive hand osteoarthritis (eHOA) is a specific type of hand osteoarthritis that occurs in fingers already affected by OA, characterized by inflammation, pain, disability, and joint erosions.
  • The study used data from the Osteoarthritis Initiative, analyzing hand radiographs and serum biomarkers over 48 months to identify participants who developed incident radiographic eHOA.
  • Results indicated that higher levels of interleukin-7, an inflammatory cytokine, are linked to the development of eHOA, suggesting it may contribute to inflammation and joint damage in affected individuals.

Objectives: There is no evidence linking specific osteoarthritis (OA) types, such as erosive hand OA (EHOA), with distant generalised changes in muscle composition (sarcopenia), which can potentially be modified. This study pioneers the exploration of the association between EHOA and sarcopenia, both of which are predominantly observed in the older adults.

Methods: Using the Osteoarthritis Initiative cohort, we selected hand OA (modified Kellgren and Lawrence (grade ≥2 in ≥1 hand joint) participants with radiographic central erosions in ≥1 joints (EHOA group) and propensity score-matched hand OA participants with no erosion (non-EHOA group).
