In this study, we report a series of α-aminobenzylphenol analogues as potent PDI inhibitors. The lead compound, , is a covalent nanomolar inhibitor of PDI, and the combination of analogues with glutathione synthesis inhibitor buthionine sulfoximine (BSO) leads to synergistic cell growth inhibition.
