  • - Pancreatic stone protein (PSP) is a new biomarker showing promise in diagnosing and predicting the outcomes of sepsis, alongside procalcitonin (PCT) and the SOFA score in a study of 285 patients.
  • - The study found that when both PSP and PCT levels were elevated, the likelihood of sepsis significantly increased (89% vs. lower percentages with a single or no biomarker increase).
  • - Although neither PSP nor PCT independently predicted clinical outcomes, PSP was noted for its utility in assessing kidney dysfunction, suggesting it can enhance clinical evaluations when used with PCT.

This study compared laboratory risk and turn-around time (TAT) between sequence-based typing (SBT) and next-generation sequencing (NGS) for human leukocyte antigen (HLA) typing. For risk assessment, we utilized the risk priority number (RPN) score based on failure mode and effect analysis (FMEA) and a risk acceptability matrix (RAM) according to the Clinical Laboratory Standards Institute (CLSI) guidelines (EP23-A). Total TAT was documented for the analytical phase, and hands-on time was defined as manual processes conducted by medical technicians.

  • The study examined the effectiveness of white blood cell (WBC)-related flags and customized cell population data (CPD) rules in detecting abnormal cells (blasts) using the Beckman Coulter DxH 900 automated hematology analyzer.
  • Out of 239 samples from patients with hematologic diseases, blasts were identified in 40 samples through manual slide review, and the combination of flags and CPD rules showed the highest sensitivity at 97.5% for blast detection.
  • The results suggest that customized CPD rules, especially when used alongside WBC-related flags, significantly enhance the detection of blasts, reducing false negatives and improving overall diagnostic accuracy.

ABO incompatibility is not considered a contraindication for hematopoietic stem cell transplantation (HSCT). We hypothesized that recipient-derived isoagglutinin (RDI) levels could play a critical role in clinical outcomes. In this study, we compared clinical outcomes such as survival, GVHD, infection, relapse, transfusion, and engraftment, among ABO-compatible patients (ABOc), ABO-incompatible patients (ABOi) with low RDI, and ABOi patients with high RDI.

  • The study focused on evaluating the T cell responses to two SARS-CoV-2 vaccines: ChAdOx1-nCoV-19 and BNT162b2, particularly since most previous research concentrated on humoral (antibody) responses.
  • Both vaccinated groups initially displayed stronger T cell responses compared to unvaccinated participants, but these responses decreased over time.
  • A BNT162b2 booster shot led to a significant increase in T cell responses, and the findings indicate that using an Interferon Gamma Release Assay (IGRA) can effectively monitor immune responses following vaccination.

  • Presepsin is an early infection marker, while KL-6 and SP-A are linked to lung infections and fibrosis; this study focused on their levels in umbilical cord blood (UCB).
  • Researchers analyzed 613 UCB samples to set reference intervals (RIs) for these markers and looked at factors like neonatal sex, gestational age, and maternal health conditions.
  • The study found specific RIs for presepsin, KL-6, and SP-A without significant differences based on the factors analyzed, though KL-6 levels slightly increased with gestational age, indicating room for further research on clinical implications.

Article Synopsis
  • Digital morphology analyzers, like the Sysmex DI-60, are increasingly used to differentiate white blood cells (WBCs) and are evaluated against traditional manual counting in lab settings, especially in cases of low WBC counts (leukopenia).
  • In a study of 40 blood samples categorized by leukopenia severity, the DI-60 demonstrated a significantly lower risk score compared to manual counting, although it took longer to process the samples.
  • The findings suggest that while DI-60 enhances safety and reduces risks in the laboratory, its use should be carefully considered based on the WBC count, particularly since it requires more time to analyze due to the lower cell counts in severe leukopenic cases.
