Background: The adoption of docetaxel for systemic treatment of metastatic prostate cancer (PCa), in both castration-sensitive (mCSPC) and castration-resistant (mCRPC) settings, is poorly understood. This study examined the real-world utilization of docetaxel in these patients and their outcomes.

Methods: A retrospective population-based study used administrative data from Ontario, Canada, to identify men aged ≥66 years who were diagnosed with de novo mCSPC or mCRPC between 2014 and 2019 and received docetaxel.

Article Synopsis
  • Autonomic neuromodulation, specifically through transcutaneous magnetic stimulation (TcMS), is being evaluated for its potential to reduce ventricular tachycardia (VT) storm episodes in patients.
  • A study involving 26 patients used a double-blind, sham-controlled design to assess the safety and efficacy of TcMS targeting the left stellate ganglion, comparing it to sham stimulation.
  • Results showed that, while freedom from VT in the first 24 hours didn't differ significantly between groups, the TcMS group experienced fewer VT episodes in the following 72 hours and required fewer antiarrhythmic drugs.

Background: Prostate cancer risk stratification using single-nucleotide polymorphisms (SNPs) demonstrates considerable promise in men of European, Asian, and African genetic ancestries, but there is still need for increased accuracy. We evaluated whether including additional SNPs in a prostate cancer polygenic hazard score (PHS) would improve associations with clinically significant prostate cancer in multi-ancestry datasets.

Methods: In total, 299 SNPs previously associated with prostate cancer were evaluated for inclusion in a new PHS, using a LASSO-regularized Cox proportional hazards model in a training dataset of 72,181 men from the PRACTICAL Consortium.

Delayed cerebral ischemia (DCI) secondary to vasospasm is a determinate of outcomes following non-traumatic subarachnoid hemorrhage (SAH). SAH patients are monitored using transcranial doppler (TCD) to measure cerebral blood flow velocities (CBFv). However, the accuracy and precision of manually acquired TCD can be operator dependent.

Behavioral language treatment approaches represent the standard of care for persons with aphasia (PWA), but the benefits of these treatments are variable. Moreover, due to the logistic and financial limitations on the amount of behavioral therapy available to patients, it is often infeasible for PWA to receive behavioral interventions with the level of frequency, intensity, or duration that would provide significant and lasting benefit, underscoring the need for novel, effective treatment approaches. Noninvasive brain stimulation (NIBS) techniques, such as transcranial magnetic stimulation (TMS) and transcranial direct current stimulation (tDCS), have emerged as promising neurally-based tools to enhance language abilities for PWA following stroke.

Article Synopsis
  • The study aims to explore differences between what cancer survivors report about their hearing loss (HL) and what audiometric tests reveal, particularly focusing on those who had cisplatin-based chemotherapy.
  • It involved 1,410 testicular cancer survivors who underwent audiometric testing and filled out questionnaires, with findings showing that 34.8% reported HL.
  • Key factors affecting their perception of HL included older age, lack of prior noise exposure, lower education levels, and the presence of tinnitus, which influenced whether they underestimated or overestimated their hearing issues.

Background: Autoimmune encephalitis (AE) is a rare collection of disorders that present with a diverse and often nebulous set of clinical symptoms. Indiscriminate use of multi-antibody panels decreases their overall utility and predictive value. Application of a standardized scoring system may help reduce the number of specimens that generate misleading or uninformative results.

Background: Right ventricular (RV) enlargement is common in endurance athletes. It is usually considered to be physiological, but it is possible that this remodelling is adverse, manifesting as a variant of arrhythmogenic right ventricular cardiomyopathy (ARVC), termed "exercise-induced ARVC." A novel biomarker (anti-desmoglein-2 [anti-DSG2] antibody) has been shown to indicate ARVC with high sensitivity and specificity and may be an immune response to breakdown of RV desmosomes.

Article Synopsis
  • Cardiovascular disease is a major issue for men with prostate cancer, and low testosterone levels can increase their risk of heart problems.
  • A study looked at 1,326 men with prostate cancer who hadn’t started hormone therapy and found that 42% of them had low testosterone.
  • Men with low testosterone were more likely to have other health issues like heart disease, diabetes, and obesity, making them at greater risk for future cardiovascular events.

Opinions about cognitive enhancement (CE) are context-dependent. Prior research has demonstrated that factors like peer pressure, the influence of authority figures, competition, moral relevance, familiarity with enhancement devices, expertise, and the domain of CE to be enhanced can influence opinions. The variability and malleability of patient, expert, and public attitudes toward CE is important to describe and predict because these attitudes can influence at-home, clinical, research, and regulatory decisions.

Biomaterials have a great potential to improve human health, however in vitro and in vivo studies are necessary to provide information on their efficacy and safety. This study reports on a comprehensive evaluation of core-shell electrospun fibers loaded with silver nanoparticles (Ag NP) where the delivery rate was controlled by different sizes of Ag NP and thermoresponsive poly(n-isopropylacrylamide) (PNIPAM) hydrogel particles. Fiber meshes also contain zinc oxide nanoparticles (ZnO NP), to improve pore structure for controlled release of Ag NP.

Article Synopsis
  • This study investigates the real-world application of treatment intensification beyond androgen-deprivation therapy (ADT) in older men with metastatic castration-sensitive prostate cancer (mCSPC) in Ontario, Canada.
  • A cohort of 3556 patients from 2014 to 2019 showed that 78.6% were treated with standard ADT, while only 11.2% received ADT plus docetaxel and 1.5% received ADT with abiraterone acetate, indicating low uptake of intensified treatment.
  • Despite evidence suggesting better outcomes with intensified therapy, most patients in the study still received only ADT, highlighting the need for improved understanding of barriers to treatment

Purpose: To determine the association between central subfield thickness (CST) variability and visual outcomes in eyes with neovascular age-related macular degeneration treated with anti-vascular endothelial growth factor therapies.

Methods: In this post hoc, treatment-agnostic analysis, patients (N = 1,752) were grouped into quartiles of increasing CST variation. The association between CST variability and best-corrected visual acuity was measured from baseline, or from the end of the loading phase, until the end of the study using a multilevel modeling for repeated-measures model.
