: The development of a five-in-one vaccine microneedle patch (five-in-one MN patch) aims to address challenges in administering vaccines against Diphtheria (DT), Tetanus (TT), Pertussis (wP), Hepatitis B (HBsAg), and type b (Hib). Combining multiple vaccines into a single patch offers a novel solution to improve vaccine accessibility, stability, and delivery efficiency, particularly in resource-limited settings. : The five-in-one MN patch consists of four distinct microneedle arrays: DT and TT vaccines are coated together on one array, while wP, HepB, and Hib vaccines are coated separately on individual arrays.

Introduction: Subthalamic nucleus deep brain stimulation (STN DBS) improves motor symptoms of Parkinson's disease (PD), but its effect on motivation is controversial. Apathy, the lack of motivation, commonly occurs in PD and is often exacerbated after surgery and its concomitant levodopa reduction. Apathy and reward processing are associated with the ventromedial prefrontal cortex (vmPFC), which standard targeting strategies avoid by targeting the dorsolateral STN.

Study Question: What is the association between infertility with or without fertility treatment and incident onset of systemic autoimmune rheumatic disease (SARD) among women who give birth?

Summary Answer: Women who experienced infertility but did not use fertility treatment had a higher incidence of SARD up to 9 years after delivery than those who did not experience infertility, even after accounting for their higher rates of preeclampsia, spontaneous preterm birth, and stillbirth.

What Is Known Already: Infertility is increasingly common and is an under-appreciated risk marker for chronic diseases in women. Despite several studies documenting abnormal immune activity in women with infertility, little is known about the association between infertility and incidence of autoimmune diseases such as SARD which disproportionately develops in reproductive-aged women.

Purpose: To analyze cartilage regeneration and clinical outcomes after human umbilical cord blood-derived mesenchymal stem cell (hUCB-MSCs) implantation on the basis of the distribution and location of medial femoral condyle (MFC) cartilage defects.

Methods: Patients who underwent hUCB-MSC implantation for MFC cartilage defects involved in isolated medial compartment osteoarthritis were included. The patients were divided into 2 groups, those with MFC defects located within the anterior-central portion (group A) and those with MFC defects extending to the posterior portion (group P).

Lung tissue-resident memory T (T) cells induced by influenza vaccination are crucial for heterosubtypic immunity upon re-exposure to the influenza virus, enabling rapid and robust responses upon reactivation. To enhance the efficacy of influenza vaccines, we induce the generation of lung T cells following intranasal vaccination with a commercial influenza vaccine adjuvanted with NexaVant (NVT), a TLR3 agonist-based adjuvant. We demonstrate that intranasal immunization with the NVT-adjuvanted vaccine provides improved protection against influenza virus infections by inducing the generation of CD4 T cells in the lungs in a type I interferon-dependent manner.

Objectives: Concern about contrast-induced acute kidney injury (CI-AKI) may delay the timely administration of contrast media for computed tomography (CT). The precise causative effect of iodinated contrast media on CI-AKI and its relevant risk factors remains an area of ongoing investigation. Therefore, this study aimed to determine the risk of CI-AKI following contrast-enhanced CT and its predisposing risk factors.

Article Synopsis
  • This study explores how a deep learning technique to reduce CT scan slice thickness from 5 mm to 1 mm can improve the detection of lung nodules by computer-aided detection (CAD) systems.
  • The retrospective analysis of 687 chest CT scans showed a significant increase in CAD performance, with the area under the receiver operating characteristic curve rising from 0.867 to 0.902 and nodule-level sensitivity improving from 0.826 to 0.916.
  • The findings suggest that using 1 mm CT scans not only enhances detection rates, particularly for smaller nodules, but also provides better clarity in identifying nodules compared to the 5 mm scans, making it beneficial for clinical practice.

Article Synopsis
  • CAR-T cells are effective against certain blood cancers but face challenges due to potential toxicity from targeting common tumor antigens that are also found on normal cells.
  • This study presents a solution using switchable CAR-T cells that utilize a tumor-targeting adaptor, allowing for targeted therapy while avoiding damage to healthy cells.
  • The approach shows promise in reducing toxicity and enhancing safety, potentially expanding the types of tumors that CAR-T therapies can effectively treat.

Background Sublobar resection for lung cancer is usually guided by cutoff values for consolidation size (maximal diameter of the solid tumor component) and consolidation-to-tumor ratio (CTR). The effects of these factors as continuous variables and the reason for established cutoffs are, to the knowledge of the authors, unexplored. Purpose To quantitatively assess the predictive value of CTR and consolidation size for cancer recurrence risk after sublobar resection in clinical stage IA lung adenocarcinoma.

  • Researchers conducted a trial comparing edoxaban monotherapy with dual antithrombotic therapy in patients with atrial fibrillation and stable coronary artery disease to assess risks of stroke and other serious outcomes.
  • The study involved 1,040 patients, with results showing fewer primary-outcome events in those on edoxaban alone (6.8% vs. 16.2% for dual therapy), indicating it may be safer.
  • Although the incidence of major bleeding was higher in the dual therapy group (14.2% vs. 4.7%), the major ischemic events were similar for both treatment groups.

[2,3-diamino--(4-(benzo[d]thiazol-2-yl)phenyl)propanamide], named as ETN101, is a novel therapeutic agent for hepatocellular carcinoma. In vitro studies examined ETN101 metabolites in human, mouse, rat, dog, and monkey hepatocytes and identified the drug-metabolizing enzymes involved using cDNA-expressed human recombinant cytochrome P450s (CYPs), carboxylesterases (CESs), -acetyltransferase (NAT) 1, and human liver cytosol. ETN101 showed similar metabolic stability across hepatocytes from five species, with particularly comparable stability in humans, rats, and monkeys.

Background: Classifying diverse skin types is crucial for promoting skin health. However, efficiently identifying and analyzing relevant biomarkers from a vast array of available genetic data is challenging. Therefore, this study aimed to develop a precise and efficient platform for analyzing specific skin biomarkers using quantitative real-time PCR (qRT-PCR) with the minimal invasive skin sampling method (MISSM).

Objective: Obesity is a key predictor of type 2 diabetes (T2D). However, metabolic complications are not solely due to increased BMI. We hypothesized that differences between genetically predicted BMI and observed BMI (BMI-diff) could reflect deviation from individual set point and may predict incident T2D.

  • This study evaluated the CXR-Age model, which uses deep learning to estimate a person's "radiographic age" based on chest X-rays, as a predictor of mortality risk in a large group of asymptomatic Asian individuals aged 50-80.
  • Researchers analyzed data from nearly 37,000 individuals over a median of 11 years, finding that a higher CXR-Age correlated with increased risk for all-cause mortality, cardiovascular, lung cancer, and respiratory disease deaths.
  • The study concluded that the CXR-Age model adds significant prognostic value beyond traditional clinical factors, indicating its potential usefulness across diverse populations.

Immunotherapy is likely the most remarkable advancement in lung cancer treatment during the past decade. Although immunotherapy provides substantial benefits, their therapeutic responses differ from those of conventional chemotherapy and targeted therapy, and some patients present unique immunotherapy response patterns that cannot be judged under the current measurement standards. Therefore, the response monitoring of immunotherapy can be challenging, such as the differentiation between real response and pseudo-response.
