Background: Metabolic syndrome heightens cardiovascular disease risk primarily through increased arterial stiffness. We previously demonstrated the involvement of YAP (Yes-associated protein) in high-fat/high-sucrose diet (HFHSD)-induced arterial stiffness via modulation of PPM1B (protein phosphatase Mg/Mn-dependent 1B)-lysine63 (K63) deubiquitination. In this study, we aimed to elucidate the role and mechanisms underlying PPM1B deubiquitination in HFHSD-induced arterial stiffness.

Microsatellite instability (MSI) detection using tumor tissue is a well-established prognostic and predictive biomarker for certain types of cancers. However, tumor tissue samples are less convenient to obtain than blood plasma samples. The main challenge facing next-generation sequencing-based MSI detection in blood plasma samples is the ultralow signal/noise ratio in plasma cell-free DNA (cfDNA).

Reduced mitochondrial quality and quantity in tumors is associated with dedifferentiation and increased malignancy. However, it remains unclear how to restore mitochondrial quantity and quality in tumors, and whether mitochondrial restoration can drive tumor differentiation. Our study shows that restoring mitochondrial function using retinoic acid (RA) to boost mitochondrial biogenesis and a mitochondrial uncoupler to enhance respiration synergistically drives neuroblastoma differentiation and inhibits proliferation.

Background: Dixon-based magnetic resonance imaging (MRI) intramuscular proton density fat fraction (PDFF) is a potentially useful imaging biomarker of muscle quality. However, multi-vendor, multi-site reproducibility of intramuscular PDFF quantification, required for large clinical studies, can be strongly dependent on acquisition and processing. The purpose of this study was (I) to develop a 6-point Dixon MRI-based acquisition and processing technique for reproducible multi-vendor, multi-site quantification of thigh intramuscular PDFF; and (II) to evaluate the ability of the technique to detect differences in thigh muscle status between operated .

Despite the fundamental role of bacterial strain variation in gut microbiota function, the number of unique strains of a species that can stably colonize the human intestine is still unknown for almost all species. Here we determine the strain richness (SR) of common gut species using thousands of sequenced bacterial isolates with paired metagenomes. We show that SR varies across species, is transferable by faecal microbiota transplantation, and is uniquely low in the gut compared with soil and lake environments.

Background: Patients with microsatellite-instability-high (MSI-H) or mismatch-repair-deficient (dMMR) metastatic colorectal cancer have poor outcomes with standard chemotherapy with or without targeted therapies. Nivolumab plus ipilimumab has shown clinical benefit in nonrandomized studies of MSI-H or dMMR metastatic colorectal cancer.

Methods: In this phase 3 open-label trial, we randomly assigned patients with unresectable or metastatic colorectal cancer and MSI-H or dMMR status according to local testing to receive, in a 2:2:1 ratio, nivolumab plus ipilimumab, nivolumab alone, or chemotherapy with or without targeted therapies.

Article Synopsis
  • The letter explores two catalysts, DAE-BPy-CoPor and Co-TAPP, in their roles for improving the oxygen-tolerant CO electroreduction reaction (CORR), particularly highlighting their activity range between -0.60 to -1.10 V under co-feeding conditions with CO and O.
  • The study utilizes grand canonical density functional theory (GC-DFT) and microkinetic simulation to determine how the hydrogen evolution reaction (HER) influences the turnover frequency (TOF) of CORR, categorizing HER effects based on positive and negative hydrogen formation energies.
  • The effects of the oxygen reduction reaction (ORR) on CORR are analyzed in three distinct scenarios, ranging from negligible influence on TOF to cases where

Article Synopsis
  • - The study focused on how various factors like temperature, sulfuric acid concentration, and different sulfate concentrations affect the transformation of lead sulfate (PbSO) into lead jarosite (Pb-J) in a sulfuric acid system.
  • - Results showed that the temperature threshold for converting PbSO to Pb-J decreased from 150 to 90 °C as sulfuric acid concentration dropped from 20 to 5 g/L, while high sulfuric acid concentration (over 40 g/L) kept PbSO stable without significant conversion.
  • - Additionally, higher concentrations of ferric iron promoted Pb-J formation, while sodium and potassium sulfates inhibited it, leading to the formation of specific jarosites (K-J and Na-J) instead

Needle-based injections currently enable the administration of a wide range of biomacromolecule therapies across the body, including the gastrointestinal tract, through recent developments in ingestible robotic devices. However, needles generally require training, sharps management and disposal, and pose challenges for autonomous ingestible systems. Here, inspired by the jetting systems of cephalopods, we have developed and evaluated microjet delivery systems that can deliver jets in axial and radial directions into tissue, making them suitable for tubular and globular segments of the gastrointestinal tract.

Positron emission tomography / computed tomography (PET/CT) plays a pivotal role in the assessment of cardiovascular diseases (CVD), particularly in the context of ischemic heart disease. Nevertheless, its application in other forms of CVD, such as infiltrative, infectious, or inflammatory conditions, remains limited. Recently, PET/CT systems with an extended axial field of view (LAFOV) have been developed, offering greater anatomical coverage and significantly enhanced PET sensitivity.

Background: Transcription factors (TFs) bind to DNA in a highly sequence-specific manner. This specificity manifests itself in vivo as differences in TF occupancy between the two alleles at heterozygous loci. Genome-scale assays such as ChIP-seq currently are limited in their power to detect allele-specific binding (ASB) both in terms of read coverage and representation of individual variants in the cell lines used.

  • Pregnane X receptor (PXR) is a protein that, when activated by various compounds, can lead to reduced effectiveness and safety of drugs by decreasing their active levels and increasing harmful byproducts.
  • To address this, drug developers need to evaluate how new drugs interact with PXR and modify them to limit any negative effects without losing their effectiveness.
  • The study describes a new approach using specialized molecules to target and degrade PXR, leading to improved anticancer effects of the chemotherapy drug paclitaxel, which is affected by PXR.

Obtaining micron-thick perovskite films of high quality is key to realizing efficient and stable positive (p)-intrinsic (i)-negative (n) perovskite solar cells, but it remains a challenge. Here we report an effective method for producing high-quality, micron-thick formamidinium-based perovskite films by forming coherent grain boundaries, in which high-Miller-index-oriented grains grow on the low-Miller-index-oriented grains in a stabilized atmosphere. The resulting micron-thick perovskite films, with enhanced grain boundaries and grains, showed stable material properties and outstanding optoelectronic performances.

Article Synopsis
  • ANGPTL3/4/8 proteins regulate lipoprotein lipase (LPL) activity, with ANGPTL8 forming complexes that influence ANGPTL4's function; however, their association with cardiovascular outcomes is under-researched.
  • This study involved measuring various ANGPTL proteins and complexes in two large population studies (LURIC and getABI) focusing on cardiovascular health and outcomes over several years.
  • Findings showed that while ANGPTL3/8 inhibited LPL activity and correlated with increased LDL-C and triglycerides, they did not predict cardiovascular death; however, ANGPTL4/8 and CD-ANGPTL4 were linked to higher diabetes prevalence and increased cardiovascular mortality risk.
