Pyruvate metabolism defects lead to severe neuropathies such as the Leigh syndrome (LS) but the molecular mechanisms underlying neuronal cell death remain poorly understood. Here, we unravel a connection between pyruvate metabolism and the regulation of the epitranscriptome that plays an essential role during brain development. Using genetically engineered mouse model and primary neuronal cells, we identify the transcription factor E4F1 as a key coordinator of AcetylCoenzyme A (AcCoA) production by the pyruvate dehydrogenase complex (PDC) and its utilization as an essential co-factor by the Elongator complex to acetylate tRNAs at the wobble position uridine 34 (U).

Article Synopsis
  • Pancreatic ductal adenocarcinoma (PDAC) shows treatment resistance due to a dense stroma and immunosuppressive microenvironment, prompting research into combining FOLFIRINOX chemotherapy with VE-822, a DNA repair inhibitor.* -
  • The study utilized PDAC spheroid models and mouse models to analyze the combination's effects on tumor growth and the immune and fibrotic environment, revealing a strong synergistic effect and increased apoptosis.* -
  • Results indicated that the FOLFIRINOX and VE-822 combo significantly inhibited tumor growth more than FOLFIRINOX alone, improved immune cell activity, and modified the tumor microenvironment, suggesting a potential strategy to enhance treatment effectiveness.*

Describing the structural complexity of seabeds is of primary importance for a number of geomorphological, hydrodynamical and ecological issues. Aiming to bring a decisive insight on the long-term development of a unified view, the present study reports on a comparative multi-site analysis of high resolution topography surveys in rough nearshore environments. The nine study sites have been selected to cover a wide variety of topographical features, including rocky and coral seabeds.

Background: This data paper describes the results of a 10-year scientific investigation of a biodiversity-rich private golf estate in south-eastern France in partnership with PatriNat (Office français de la biodiversité/Centre national de la recherche scientifique/Muséum national d'Histoire naturelle, Institut de Recherche pour le Développement). In total, 3,160 species and subspecies, including 1,796 arthropods and 1,049 flora, were inventoried and 65 habitat types were surveyed and mapped. This project is the first All taxa biodiversity inventory (ATBI) in a private property in France with all information available in open data.

Article Synopsis
  • The MEDITREME trial tested the safety and efficacy of a combination treatment of durvalumab, tremelimumab, and mFOLFOX6 chemotherapy in patients with RAS-mutant unresectable metastatic colorectal cancer (CRC).
  • The study found no safety issues and met its primary efficacy goal with a 90.7% progression-free survival (PFS) rate at three months for patients with microsatellite stable tumors.
  • Overall, responders showed higher tumor mutational burden and better outcomes linked to immune activity, indicating that this treatment combination is promising for managing MSS mCRC.
