Three central steps during the biosynthesis of cytochalasan precursors, including reductive release, Knoevenagel cyclisation and Diels Alder cyclisation are not yet understood at a detailed molecular level. In this work we investigated the reductive release step catalysed by a hybrid polyketide synthase non-ribosomal peptide synthetase (PKS-NRPS) from the pyrichalasin H pathway. Synthetic thiolesters were used as substrate mimics for in vitro studies with the isolated reduction (R) and holo-thiolation (T) domains of the PKS-NRPS hybrid PyiS.

Submerged cultivation of MUCL 54604 resulted in formation of a stereoisomeric mixture of new sulfur-containing sporothriolide derivatives named sporothioethers A and B. The presence of the 2-hydroxy-3-mercaptopropanoic acid moiety attenuates the antimicrobial activity in comparison to the precursor sporothriolide suggesting a detoxification mechanism. However, moderate effects on biofilms of and were observed for sporothriolide and sporothioethers A and B at concentrations below their MICs.

Epistemic curiosity as the desire to acquire new knowledge and ideas is considered as an important attribute for successful entrepreneurs among practitioners, yet there is lacking empirical evidence of epistemic curiosity having an effect on entrepreneurial outcomes. This study aims to put a spotlight on epistemic curiosity as a predictor for entrepreneurial intentions and orientation. We found that epistemic curiosity has a stronger influence on entrepreneurial outcomes in comparison to the Big Five personality , which is a widely used and conceptually related predictor for entrepreneurship.

Instabilities and fluctuations of the plasma jet in a thermal spray process can have a significant influence on the particle in-flight temperatures and velocities, affecting the properties of resulting plasma-sprayed coatings. Presented in this paper is a novel method for capturing the effects particles are exposed to in the plasma spraying process. High-speed camera images of a plasma jet generated by a cascaded three-cathode plasma generator (TriplexPro-210) were recorded for varying operating conditions.
