Surgical resection or hypo-fractionated radiation therapy (RT) in early-stage non-small cell lung cancer (NSCLC) achieves local tumor control, but metastatic relapse remains a challenge. We hypothesized that immunotherapy with anti-CTLA-4 and bempegaldesleukin (BEMPEG; NKTR-214), a CD122-preferential IL2 pathway agonist, after primary tumor RT or resection would reduce metastases in a syngeneic murine NSCLC model. In our model of protein malnutrition (R21 group), bronchus-associated lymphoid tissue (BALT), situated in the lower respiratory tract, showed a decrease of CD4(+), CD8alpha(+), and TCRalphabeta(+) lymphocytes but TCRgammadelta(+) cells were increased. CD5+ and CD4+ lymphocytes gradually increased with age. A predominance of CD8alpha+ over CD4+ T cells was found from 7 to 45 days but from 45 to 60 days of age T helper cells outnumbered the cytotoxic subpopulation.
