In the binding process between proteins and ligand molecules, water molecules play a pivotal role by forming hydrogen bonds that enable proteins and ligand molecules to bind more strongly. However, current methodologies for predicting binding affinity overlook the importance of water molecules. Therefore, we developed a model called GraphWater-Net, specifically designed for predicting protein-ligand binding affinity, by incorporating water molecules.

Nylon 56/cotton (N/C) blended fabrics have excellent durability and can cater to various clothing needs. However, the quality and value of textile garments are significantly influenced by the dyeing process. Due to variations in the structure and characteristics of nylon 56 and cotton fibers in blended fabrics, achieving consistent coloration is challenging, resulting in noticeable color discrepancies.

Article Synopsis
  • - Cannabidiol (CBD), a non-psychoactive compound from cannabis, shows potential in treating various central nervous system disorders like epilepsy and chronic pain, but its exact pharmacological mechanisms remain unclear.
  • - This study identified the transient receptor potential canonical 4 (TRPC4) channel as a new target for CBD, demonstrating that CBD can inhibit TRPC4, affecting neuronal excitability in the medial prefrontal cortex.
  • - Using whole-cell electrophysiology, researchers found that CBD inhibits the TRPC4 channel's activity, particularly in response to epinephrine, suggesting CBD may reduce neuron excitability by acting on TRPC4 channels.

In newly diagnosed multiple myeloma (NDMM), measurable residual disease (MRD) status is prognostically important, but its role in treatment-decisions remains unclear. In a phase II trial (NCT04113018), we assessed daratumumab, carfilzomib, lenalidomide and dexamethasone (Dara-KRd) induction followed by a next generation sequencing (NGS) based MRD-adapted strategy. The primary endpoint was rate of complete response and stringent complete response (≥CR) after induction.

A novel magnetorheological vibration isolator with two operating conditions, horizontal shear and vertical compression, was designed and manufactured. The rheological properties of the energy-dissipating material were directly related to the volume fraction of iron powder in the laminated working unit of the magnetorheological vibration isolator. Aggregation of the carbonyl iron powder (CIP) strongly influenced on the rheological properties of the magnetorheological vibration isolator.

Although bioinformatics-based methods accurately identify SEs (Super-enhancers), the results depend on feature design. It is foundational to representing biological sequences and automatically extracting their key features for improving SE identification. We propose a deep learning model MuSE (Multi-Feature Fusion for Super-Enhancer), based on multi-feature fusion.

Immunotherapy has emerged as a vital component in the contemporary landscape of cancer treatment. Recent studies have indicated that CEP55 plays an oncogenic role; however, its specific mechanisms in promoting tumor proliferation and its potential value in prognosis and immunotherapy prediction across various cancers remain to be elucidated. CEP55 was significantly overexpressed in 22 cancer types compared with their adjacent normal tissues.

The subcellular localization of circular RNAs (circRNAs) is crucial for understanding their functional relevance and regulatory mechanisms. CircRNA subcellular localization exhibits variations across different cell lines, demonstrating the diversity and complexity of circRNA regulation within distinct cellular contexts. However, existing computational methods for predicting circRNA subcellular localization often ignore the importance of cell line specificity and instead train a general model on aggregated data from all cell lines.

γ-Aminobutyric acid (GABA) neurotransmission alterations have been implicated to play a role in depression pathogenesis. While GABA receptor positive allosteric modulators are emerging as promising in clinical practice, their precise antidepressant mechanism remains to be further elucidated. The aim of the present study was to investigate the effects of LY-02, a novel compound derived from the metabolite of timosaponin, on depression in animals and its mechanism.

  • Identifying early events in neurodegenerative disorders, like Huntington's disease (HD), is essential for creating preventive treatments, particularly focusing on the role of dysfunctional indirect pathway spiny projection neurons (iSPNs) and increased dopamine levels.
  • The study reveals that genetic disruption of iSPN function in mice leads to heightened levels of striatal dopamine, potentially causing early symptoms like hyperkinesia, before observable dysfunction occurs.
  • By analyzing iSPNs, researchers found that reducing the protein GSTO2 could prevent dopaminergic issues and delay hyperkinetic symptoms, highlighting the significance of maintaining dopamine balance in HD progression.

  • China is facing a growing population of older adults, which increases the risk of developing tuberculous pleural effusion (TPE) due to weakened immune systems with age.
  • A study analyzed 519 patients aged 60 and above to determine the effectiveness of seven biomarkers in diagnosing TPE, identifying Effusion ADA, IGRA, and Effusion LDH/ADA as key differentiators between TPE and non-TPE.
  • The combination of these biomarkers achieved high diagnostic accuracy for TPE in older patients, with an AUC of 0.925, sensitivity of 85.23%, and specificity of 89.57%, emphasizing the importance of multiple indicators for effective diagnosis.

A sandwich-type electrochemical aptasensor for ultrasensitive detection of glypican-3 (GPC3) was constructed using GPC3 aptamer (GPC3) labelled reduced graphene oxide-cerium oxide-gold nanoparticles (RGO-CeO-Au NPs) as the signal probe and the same GPC3 as the capture probe. The electrochemical redox properties of CeO (Ce/Ce) in the RGO-CeO-Au NPs indicate the electrochemical signals. When the target GPC3 was present, an "aptamer-protein-aptamer" sandwich structure was formed on the sensing interface due to the specific binding between the protein and aptamers, resulting in an increased electrochemical redox signal detected by differential pulse voltammetry (DPV) technique.

Developing aggregation-induced emission (AIE)-based hydrogels that exhibit fluorescence enhancement as to thermal properties is an interesting and challenging task. In this work, we employed the fluorophore 2'-hydroxychalcone (HC), fluorescence properties of which are easily influenced by the excited-state intramolecular proton transfer and twisted intramolecular charge transfer (TICT) effects, to develop a novel type of temperature-sensitive polymers, hydroxychalcone-based polymers (HCPs). By controlling the temperature-dependent water microenvironments in HCPs, the intramolecular hydrogen bonds between water and HCPs can be regulated, thereby influencing the TICT process and leading to thermo-induced fluorescence enhancement, which shows a contrary tendency compared to typical AIEgens that always exhibit fluorescence attenuation as the thermal energy accelerates the molecular motion.
