Article Synopsis
  • Recent research on immunotherapy for Alzheimer's disease highlights the need to understand how different antibodies bind to amyloid-beta (Aβ), as this affects their effectiveness and potential side effects.
  • Lecanemab, a specific IgG1 antibody, was studied using brain samples from Alzheimer's patients and non-demented controls to explore its interactions with various Aβ forms, revealing high levels of insoluble Aβ, especially Aβ42, in Alzheimer's subjects.
  • Findings indicate that lecanemab binds primarily to soluble aggregated Aβ protofibrils rather than insoluble Aβ plaques, suggesting a targeted approach in treating Alzheimer's through the modulation of these specific proteins.

Background and purpose - Swedish clinical guidelines for osteoarthritis (OA) prioritize patient education, exercise, and-if necessary-weight reduction before considering adjunct pharmacological intervention. Contrariwise, we investigated the proportion and type of dispensed analgesic prescriptions in Sweden received by patients during 3 years before commencing non-pharmacological primary care interventions for OA (2008-2016) compared with the general population. Furthermore, we analyzed the proportion of analgesic prescriptions dispensed before (2008-2012) compared with after (2012-2016) guideline publication in terms of concordance with clinical guideline recommendations.

Background: National guidelines are being produced at an increasing rate, and politicians and managers are expected to promote these guidelines and their implementation in clinical work. However, research seldom deals with how decision-makers can perceive these guidelines or their challenges in a cultural context. Therefore, the aim of this study was twofold: to investigate how well Promoting Action on Research Implementation in Health Services (PARIHS) reflected the empirical reality of mental healthcare and to gain an extended understanding of the perceptions of decision-makers operating within this context, in regard to the implementation of evidence-based guidelines.

Article Synopsis
  • A study investigated the prevalence of kinesiophobia (fear of movement) in patients who underwent surgery for lumbar disc herniation, which had not been previously explored.
  • Out of 97 patients surveyed 10-34 months post-surgery, 36 reported kinesiophobia, and this was linked to worse overall outcomes in terms of disability, pain, and mental health measures.
  • The findings suggest that kinesiophobia is common after disc surgery and negatively impacts recovery and quality of life for those affected.

Objective: NXY-059 is a novel free-radical trapping neuroprotectant. Digoxin treatment is common in acute ischaemic stroke, the intended patient population for NXY-059. Since both digoxin and NXY-059 are eliminated primarily renally, with a substantial contribution by active renal secretion, and because digoxin has a narrow therapeutic window, this open, randomised, crossover, two-period study investigated whether NXY-059 affects the pharmacokinetics (PK) of digoxin.

Two hundred twenty-nine consecutive isolates of Mycobacterium tuberculosis complex from patients with pulmonary tuberculosis in Guinea-Bissau, which is located in West Africa, were analyzed for clonal origin by biochemical typing and DNA fingerprinting. By using four biochemical tests (resistance to thiophene-2-carboxylic acid hydrazide, niacin production, nitrate reductase test, and pyrazinamidase test), the isolates could be assigned to five different biovars. The characteristics of four strains conformed fully with the biochemical criteria for M.

Human dUTPase, expressed in Escherichia coli, has been crystallized. Single crystals were obtained by the vapour-diffusion technique using 2-propanol and PEG 4000 as precipitants. The enzyme was co-crystallized with the substrate dUTP and a metal chelator EDTA to prevent hydrolysis of the substrate.
