In this open study, the therapeutic effect of moclobemide, a reversible selective monoamine oxidase A inhibitor, was tested in 20 patients with Parkinson's disease who developed levodopa-induced motor response complications. Moclobemide as adjunct therapy reduced "off" time duration for 27%, without an overall motor and functional improvement during their "on" periods. Since it was well tolerated, moclobemide may be specially indicated in elderly or depressed fluctuating parkinsonian patients.

View Article and Find Full Text PDFChanges in silent period (SP) duration following transcranial magnetic stimulation (TMS) set at 20% above the motor threshold were studied in six subjects suffering from writer's cramp, while performing dystonic movement and during voluntary isometric contraction of the muscles mostly involved in the dystonic movement. Dependency of SP duration on the intensity of preceding muscle contraction was compared on both affected and healthy side. In all subjects SP duration during dystonic contraction was shorter than during voluntary contraction of the similar strength performed with the same hand.

Background: Symptomatic (secondary) dystonias associated isolated lesions in the brain provide insight into etiopathogenesis of the idiopathic form of dystonia and are a basis for establishing the possible correlation between the anatomy of a lesion and the type of dystonia according to muscles affected.
Methods: In 358 patients with differently distributed dystonias, a group of 16 patients (4.5%) was encountered in whom dystonia was associated with focal brain lesions.
Six of 13 cloned members of the human tRNA(Val) gene family code for tRNA(Val) pseudogenes, of which all but one are transcribed efficiently in HeLa cell extracts. Due to single or multiple mismatches in stem regions, the corresponding pre-tRNAs are resistant against the action of human 5'- and 3'-processing enzymes and are thus prevented from being converted to mature tRNAs. Surprisingly, all of them are accurately and efficiently processed to mature-sized tRNA in yeast nuclear extract.

Two tRNA(Val) pseudogenes and a tRNA(Val) variant gene were found to be associated with tRNA genes encoding tRNA(ArgICG), tRNA(GlyUCC), and tRNA(ThrIGU), respectively, on distinct DNA fragments. All tRNA genes, including the pseudogenes, are actively transcribed in HeLa nuclear extract.

Two years prior the patient suffered from acute abdomen and underwent apendectomy with slow wound healing and a stomach edema 6 months later with a finding of smaller changes on the left side of the thorax. The patient was hospitalized when he already had an advanced stage of the disease.
