2-Oxoglutarate (2OG) dependent N-methyl lysine demethylases (JmjC-KDMs) regulate eukaryotic transcription. We report studies showing that isolated forms of all human KDM4 and KDM5 JmjC enzymes catalyse demethylation of N-methylated Arg-3 of histone H2a. Unexpectedly, the results reveal that KDM4E and, less efficiently, KDM4D catalyse C-4 hydroxylation of Arg-20 of H2a on peptides, recombinant H2a, and calf histone extracts, including when the Arg-20 guanidino group is N-methylated.

Article Synopsis
  • - The HALP score, which assesses levels of hemoglobin, albumin, lymphocytes, and platelets, was evaluated in patients with acute ischemic stroke (AIS) who underwent mechanical thrombectomy to predict 90-day mortality.
  • - In a study of 122 patients, a HALP score below 13.10 was linked to an increased risk of death and was associated with more cases of large artery thrombosis, while higher scores were correlated with cardioembolism and hyperlipidemia.
  • - The HALP score is a simple and cost-effective way to assess mortality risk in AIS patients, indicating that lower scores reflect worse prognoses and highlighting its relevance in survival predictions during reperfusion therapy.
