Here we describe a general strategy to engineer large (more than 1 Mbp) focal amplifications mediated by extrachromosomal DNAs (ecDNAs) in a spatiotemporally controlled manner in cells and in mice. By coupling ecDNA formation with expression of selectable markers, we track the dynamics of ecDNA-containing cells under physiological conditions and in the presence of specific selective pressures.

Introduction: The outcome of thalamic deep brain stimulation (DBS) for essential tremor (ET) varies, probably due to the difficulty in identifying the optimal target for DBS placement. Recent approaches compared the clinical response with a connectivity-based segmentation of the target area. However, studies are contradictory by indicating the connectivity to the primary motor cortex (M1) or to the premotor/supplementary motor cortex (SMA) to be therapeutically relevant.

Perioperative Observations and Outcome in Surgical Treatment of Malignant Peripheral Nerve Sheath Tumors.

Background/objectives: This retrospective observational study aimed to investigate the perioperative outcome in Malignant Peripheral Nerve Sheath Tumors (MPNSTs) with and without relation to Neurofibromatosis Type 1 (NF1) and to detect possible influencing factors.

Methods: Clinical reports, histopathological evaluations, imaging, and treatment characteristics were reviewed in 35 operated MPNSTs in 33 patients. Possible predictive valuables included disease type, preoperative tumor volume, SUV and MIB-1 proliferation index, resection margins, the presence of metastasis, and whether radio-/chemotherapy was received.

The combination of solid oxide fuel cells (SOFCs) and wood gasification has the potential to significantly increase renewable electricity production and decrease emissions. Depending on the quality of the wood gas, degradation processes have a significant impact on the reliability and lifetime of the SOFC. Using electrochemical impedance spectroscopy (EIS) and subsequent distribution of relaxation times (DRT) analysis, the impact on the degradation of coupling wood gasification with a commercial SOFC stack is determined in this study.

Polarization-resolved mode evolution in TMI-limited Yb-doped fiber amplifiers using a novel high-speed Stokes polarimeter.

In this work we have developed a high-speed Stokes polarimeter method based on simultaneous 4-channel imaging with a high-speed camera. Thus, current speed limitations of imaging polarimeters for wavelengths around 1 µm can be overcome, allowing a sub-ms polarization-resolved characterization of transverse mode instability (TMI). Additionally, the Stokes parameters of each individual mode are calculated by a simultaneous 4-beam mode reconstruction algorithm during post-processing and can be analyzed with unprecedented temporal resolution.

Integrative Chemical-Biological Grouping of Complex High Production Volume Substances from Lower Olefin Manufacturing Streams.

Human cell-based test methods can be used to evaluate potential hazards of mixtures and products of petroleum refining ("unknown or variable composition, complex reaction products, or biological materials" substances, UVCBs). Analyses of bioactivity and detailed chemical characterization of petroleum UVCBs were used separately for grouping these substances; a combination of the approaches has not been undertaken. Therefore, we used a case example of representative high production volume categories of petroleum UVCBs, 25 lower olefin substances from low benzene naphtha and resin oils categories, to determine whether existing manufacturing-based category grouping can be supported.

We present a case that illustrates the complex contexts in which forensic veterinary pathology examinations may be carried out. A wild muskrat (Ondatra zibethicus) had died after a putative bite attack from a domestic dog. Bite attacks by privately owned dogs on wild animals in Switzerland violate the Swiss civil (and/or hunting) laws, and dog owners are generally punished with a monetary fine; hence, this case appeared to be straightforward.

Xenosurveillance proof-of-principle: Detection of and SARS-CoV-2 antibodies in mosquito blood meals by (pan)-specific ELISAs.

Blood-fed insects can be used to analyse the host blood for circulating vertebrate pathogens or antibodies directed against them. We tested whether naturally acquired antibodies in different host species can be detected by host-specific and pan-specific ELISAs in mosquito blood meals. Cat- and alpaca-specific ELISAs could detect antibodies against or SARS-CoV-2 in blood meals of for 48 and at least 24 h, respectively.
