Cyclooxygenase-2 (COX-2) is present in a healthy brain at low densities but can be markedly upregulated by excitatory input and by inflammogens. This study evaluated the sensitivity of the PET radioligand [C]-6-methoxy-2-(4-(methylsulfonyl)phenyl)--(thiophen-2-ylmethyl)pyrimidin-4-amine ([C]MC1) to detect COX-2 density in a healthy human brain. The specificity of [C]MC1 was confirmed using lipopolysaccharide-injected rats and transgenic mice expressing the human gene, with 120-min baseline and blocked scans using COX-1 and COX-2 selective agents.

Article Synopsis
  • Recent autopsy studies show that interface astroglial scarring (IAS) can occur at the gray-white matter junction in military personnel who experience repeated blast brain injuries.
  • There is currently no neuroimaging test available to detect IAS, making it difficult to diagnose and treat these injuries.
  • In a study of 27 U.S. Special Operations Forces personnel, five individuals (18.5%) showed elevated neuroinflammation signals at the gray-white matter interface compared to healthy controls, suggesting that TSPO PET scans may help identify repeated blast brain injury.

Objectives: The study aimed to analyse outcomes of surgery for blood culture-negative infective endocarditis (BCNIE) and to evaluate the role of molecular biological imaging.

Methods: Patients undergoing surgery for native or prosthetic valve endocarditis from 2013 to 2022 were analysed regarding blood culture-positive infective endocarditis (BCPIE) and BCNIE. For laboratory diagnostics in BCNIE, excised valves or prostheses underwent conventional microbiological culture and fluorescence in situ hybridization combined with 16S rRNA-gene polymerase chain reaction and sequencing (FISHseq).

Our laboratory recently developed [C]PS13 as a PET radioligand to selectively measure cyclooxygenase-1 (COX-1). The cyclooxygenase enzyme family converts arachidonic acid into prostaglandins and thromboxanes, which mediate inflammation. The total brain uptake of [C]PS13, which is composed of both specific binding and background uptake, can be accurately quantified with gold standard methods of compartmental modeling.

Functional Positron Emission Tomography (fPET) with (bolus plus) constant infusion of [F]-fluorodeoxyglucose FDG), known as fPET-FDG, is a recently introduced technique in human neuroimaging, enabling the detection of dynamic glucose metabolism changes within a single scan. However, the statistical analysis of fPET-FDG data remains challenging because its signal and noise characteristics differ from both classic bolus-administration FDG PET and from functional Magnetic Resonance Imaging (fMRI), which together compose the primary sources of inspiration for analytical methods used by fPET-FDG researchers. In this study, we present an investigate of how inaccuracies in modeling baseline FDG uptake can introduce artifactual patterns to detrended TAC residuals, potentially introducing spurious (de)activations to general linear model (GLM) analyses.

Segmentation of brain structures on magnetic resonance imaging (MRI) is a highly relevant neuroimaging topic, as it is a prerequisite for different analyses such as volumetry or shape analysis. Automated segmentation facilitates the study of brain structures in larger cohorts when compared with manual segmentation, which is time-consuming. However, the development of most automated methods relies on large and manually annotated datasets, which limits the generalizability of these methods.

Affine image registration is a cornerstone of medical-image analysis. While classical algorithms can achieve excellent accuracy, they solve a time-consuming optimization for every image pair. Deep-learning (DL) methods learn a function that maps an image pair to an output transform.

De-identification of medical images intended for research is a core requirement for data sharing initiatives, particularly as the demand for data for artificial intelligence (AI) applications grows. The Center for Biomedical Informatics and Information Technology (CBIIT) of the United States National Cancer Institute (NCI) convened a two half-day virtual workshop with the intent of summarizing the state of the art in de-identification technology and processes and exploring interesting aspects of the subject. This paper summarizes the highlights of the second day of the workshop, the recordings and presentations of which are publicly available for review.

Introduction: The human cerebellum emerges as a posterior brain structure integrating neural networks for sensorimotor, cognitive, and emotional processing across the lifespan. Developmental studies of the cerebellar anatomy and function are scant. We examine age-dependent MRI morphometry of the anterior cerebellar vermis, lobules I-V and posterior neocortical lobules VI-VII and their relationship to sensorimotor and cognitive functions.

  • Consciousness has two main parts: being awake (arousal) and being aware of things.
  • Scientists are trying to learn more about how the brain keeps us awake and alert, focusing on parts below the brain's surface.
  • They found important brain areas that help with wakefulness and connected them to parts that help with awareness, showing how these different brain networks work together.

  • The effects of repeated blast exposure (RBE) on the brain health of US Special Operations Forces (SOF) are not fully understood, and currently, there is no test to diagnose injury from such exposures.
  • A study involving 30 active-duty US SOF found that higher blast exposure correlates with changes in brain structure and cognitive performance, particularly affecting the rostral anterior cingulate cortex (rACC).
  • These findings indicate that increased blast exposure can lead to health-related issues and suggest that a comprehensive, network-based diagnostic method may be beneficial for identifying brain injuries in SOF personnel.

We briefly review recent advances in the hydrodynamic modeling of the dynamics of droplets on adaptive substrates, in particular, solids that are covered by polymer brushes. Thereby, the focus is on long-wave and full-curvature variants of mesoscopic hydrodynamic models in gradient dynamics form. After introducing the approach for films/drops of nonvolatile simple liquids on a rigid smooth solid substrate, it is first expanded to an arbitrary number of coupled degrees of freedom before considering the specific case of drops of volatile liquids on brush-covered solids.

Passive surface acoustic wave (SAW) devices are attractive candidates for continuous wireless monitoring of corrosion in large infrastructures. However, acoustic loss in the aqueous medium and limited read range usually create challenges in their widespread use for monitoring large systems such as oil and gas (O&G) pipelines, aircraft, and processing plants. This paper presents the investigation of impedance-loaded reflective delay line (IL-RDL) SAW devices for monitoring metal corrosion under O&G pipeline-relevant conditions.

Several cardiovascular and metabolic indicators, such as cholesterol and blood pressure have been associated with altered neural and cognitive health as well as increased risk of dementia and Alzheimer's disease in later life. In this cross-sectional study, we examined how an aggregate index of cardiovascular and metabolic risk factor measures was associated with correlation-based estimates of resting-state functional connectivity (FC) across a broad adult age-span (36-90+ years) from 930 volunteers in the Human Connectome Project Aging (HCP-A). Increased (i.

We study the static and dynamic wetting of adaptive substrates using a mesoscopic hydrodynamic model for a liquid droplet on a solid substrate covered by a polymer brush. First, we show that on the macroscale Young's law still holds for the equilibrium contact angle and that on the mesoscale a Neumann-type law governs the shape of the wetting ridge. Following an analytic and numeric assessment of the static profiles of droplet and wetting ridge, we examine the dynamics of the wetting ridge for a liquid meniscus that is advanced at constant mean speed.

Mesh-based reconstruction of the cerebral cortex is a fundamental component in brain image analysis. Classical, iterative pipelines for cortical modeling are robust but often time-consuming, mostly due to expensive procedures that involve topology correction and spherical mapping. Recent attempts to address reconstruction with machine learning methods have accelerated some components in these pipelines, but these methods still require slow processing steps to enforce topological constraints that comply with known anatomical structure.

In this paper we describe and validate a longitudinal method for whole-brain segmentation of longitudinal MRI scans. It builds upon an existing whole-brain segmentation method that can handle multi-contrast data and robustly analyze images with white matter lesions. This method is here extended with subject-specific latent variables that encourage temporal consistency between its segmentation results, enabling it to better track subtle morphological changes in dozens of neuroanatomical structures and white matter lesions.
