To successfully apply the genome editing technology using the CRISPR/Cas9 system in the clinic, it is necessary to achieve a high efficiency of knock-in, which is insertion of a genetic construct into a given locus of the target cell genome. One of the approaches to increase the efficiency of knock-in is to modify donor DNA with the same Cas9 targeting sites (CTS) that are used to induce double-strand breaks (DSBs) in the cell genome (the double-cut donor method). Another approach is based on introducing truncated CTS (tCTS), including a PAM site and 16 proximal nucleotides, into the donor DNA.

The low knock-in efficiency, especially in primary human cells, limits the use of the genome editing technology for therapeutic purposes, rendering it important to develop approaches for increasing the knock-in levels. In this work, the efficiencies of several approaches were studied using a model of knock-in of a construct coding for the peptide HIV fusion inhibitor MT-C34 into the human CXCR4 locus in the CEM/R5 T cell line. First, donor DNA modification was evaluated as a means to improve the efficiency of plasmid transport into the nucleus.

Synchronization of Bloch oscillations in small Josephson junctions (JJs) under microwave radiation, which leads to current quantization, has been proposed as an effect that is dual to the appearance of Shapiro steps. This current quantization was recently demonstrated in superconducting nanowires in a compact high-impedance environment. Direct observation of current quantization in JJs would confirm the synchronization of Bloch oscillations with microwaves and help with the realisation of the metrological current standard.

We apply the analytically solvable model of two electrons in two orbitals to diradical molecules, characterized by two unpaired electrons. The effect of doubly occupied and empty orbitals is taken into account by means of random phase approximation (RPA). We show that in the static limit, the direct RPA leads to the renormalization of the parameters of the two-orbital model.

The fragile nature of quantum circuits is a major bottleneck to scalable quantum applications. Operating at cryogenic temperatures, quantum circuits are highly vulnerable to amplifier backaction and external noise. Non-reciprocal microwave devices such as circulators and isolators are used for this purpose.

Gene editing using the CRISPR/Cas9 system provides new opportunities to treat human diseases. Approaches aimed at increasing the efficiency of genome editing are therefore important to develop. To increase the level of editing of the CXCR4 locus, which is a target for gene therapy of HIV infection, the Cas9 protein was modified by introducing additional NLS signals and ribonucleoprotein complexes of Cas9 and guide RNA were stabilized with poly-L-glutamic acid.

The Josephson junction is a building block of quantum circuits. Its behavior, well understood when treated as an isolated entity, is strongly affected by coupling to an electromagnetic environment. In 1983, Schmid predicted that a Josephson junction shunted by a resistance exceeding the resistance quantum R = h/4e ≈ 6.

The transcriptional coactivators Yes-associated protein (YAP) and transcriptional coactivator with PDZ-binding motif (TAZ) are the main downstream effectors of the evolutionarily conserved Hippo signaling pathway. YAP/TAZ are implicated in the transcriptional regulation of target genes that are involved in a wide range of key biological processes affecting tissue homeostasis and play dual roles in the aging process, depending on the cellular and tissue context. The aim of the present study was to investigate whether pharmacological inhibitors of Yap/Taz increase the lifespan of .

Electrons in two different normal metallic electrodes attached to a sufficiently thin superconducting island may become entangled due to the effect of Cooper pair splitting. This phenomenon is of fundamental importance and may also have serious implications for developing quantum communication technologies. One way to identify Cooper pair splitting is to analyze long-range cross correlations of fluctuating currents in three-terminal hybrid normal-superconducting-normal nanostructures.

We measure the rates and coupling coefficients for local Andreev, nonlocal Andreev, and elastic cotunneling processes. The nonlocal Andreev process, giving rise to Cooper pair splitting, exhibits the same coupling coefficient as the elastic cotunneling whereas the local Andreev process is more than 2 orders of magnitude stronger than the corresponding nonlocal one. Theory estimates describe the findings and explain the large difference in the nonlocal and local coupling arising from competition between electron diffusion in the superconductor and tunnel junction transparency.

Heat is detrimental for the operation of quantum systems, yet it fundamentally behaves according to quantum mechanics, being phase coherent and universally quantum-limited regardless of its carriers. Due to their robustness, superconducting circuits integrating dissipative elements are ideal candidates to emulate many-body phenomena in quantum heat transport, hitherto scarcely explored experimentally. However, their ability to tackle the underlying full physical richness is severely hindered by the exclusive use of a magnetic flux as a control parameter and requires complementary approaches.
