Commutability is where the measurement response for a reference material (RM) is the same as for an individual patient sample with the same concentration of analyte measured using two or more measurement systems. Assessment of commutability is essential when the RM is used in a calibration hierarchy or to ensure that clinical measurements are comparable across different measurement procedures and at different times. The commutability of three new Standard Reference Materials (SRMs) for determining serum total 25-hydroxyvitamin D [25(OH)D], defined as the sum of 25-hydroxyvitamin D [25(OH)D] and 25-hydroxyvitamin D [25(OH)D], was assessed through an interlaboratory study.

Background: Pathogenic variants in may lead to a syndromic genetic aortopathy. Heritable thoracic aortic disease (HTAD) and arterial events may occur in -related disease but there are limited outcomes data on vascular events in this condition.

Methods: Clinical data, phenotypical features and aortic outcomes in individuals with pathogenic/likely pathogenic (P/LP) variants enrolled in the Montalcino Aortic Consortium registry were reviewed.

The study aimed to propose sensitive, specific and easy pregnancy diagnosis for American Bison. Trans-rectal Palpation and Ultrasonography (TRPUS), concentration of Pregnancy Associated Glycoproteins (PAG), Progesterone (P4), Estrone (E1), Estrone-Sulfate (E1S) in serum and P4 and E1S in feces were compared. Immunoassay was used for PAG, whereas steroids were assayed using Liquid Chromatography coupled to Mass Spectrometry.

Bones are now recognised as endocrine organs with diverse functions. Osteocalcin, a protein primarily produced by osteoblasts, has garnered significant attention. Research into osteocalcin has revealed its impact on glucose metabolism and its unexpected endocrine role, particularly in its undercarboxylated form (ucOC).

  • The aging population has sparked increased interest in monitoring skeletal diseases like osteoporosis, leading to advancements in bone turnover biomarkers since the 2000s.
  • Various markers from type I collagen, such as CTX, NTX, PINP, and ICTP, have been developed, but their complex structures make them difficult to analyze.
  • This study aims to characterize plasma and serum CTX by directly extracting type I collagen peptides from human samples using advanced proteomics techniques, resulting in the identification of multiple CTX species for better understanding of these biomarkers.

Article Synopsis
  • The study investigates the vitamin D metabolite ratio (VMR) as a potential better indicator of vitamin D deficiency compared to 25-hydroxyvitamin D (25(OH)D) and 24,25-dihydroxyvitamin D (24,25(OH)D), aiming to show its stronger correlation with parathyroid hormone (PTH) levels and mortality.
  • It analyzed serum samples from 204 older adults, demonstrating that VMR had the strongest negative correlation with PTH levels and was associated with higher mortality rates in patients with low levels.
  • The results suggest that VMR could be a more reliable biomarker for assessing functional vitamin D deficiency, supporting its potential use in clinical practice.

In order to improve our healthcare system, it is undeniable that the future of modern medicine must focus on a more preventive and personalized approach, notably based on the individual characteristics specific to each patient. In this perspective, clinical metabolomics, which focuses on metabolites, emerges as a particularly interesting and promising approach. Indeed, this science reflects the internal and external stimuli received by an individual, thus capturing their physiological and/or pathological state.

Background: Marfan syndrome (MFS) is a multisystem disease with a unique combination of skeletal, cardiovascular and ocular features. Geleophysic/acromicric dysplasias (GPHYSD/ACMICD), characterised by short stature and extremities, are described as 'the mirror image' of MFS. The numerous pathogenic variants identified in MFS are located all along the gene and lead to the same final pathogenic sequence.

  • DNA-based vaccines, such as PlaCCine, show promise as a safe and stable immunization method, using a synthetic DNA delivery system that is not reliant on viruses or devices.
  • The efficacy of the vaccines was demonstrated in mice, with significant improvements in spike-specific humoral and cellular immune responses following intramuscular injections of variants pVAC15, pVAC16, and pVAC17, maintaining these responses for up to 14 months.
  • In studies involving K18 hACE2 transgenic mice, these DNA vaccines resulted in over a 90% reduction in lung viral loads and improved clinical outcomes, suggesting their potential for further development against SARS-CoV-2 variants.

Cardiovascular diseases have cast a significant negative impact on the lives of millions worldwide. Over the years, extensive efforts have been dedicated to enhancing diagnostic and prognostic tools for these diseases. A growing body of evidence indicates that the angiotensin convertase enzyme (ACE) and the angiotensin convertase enzyme 2 (ACE2), and angiotensin peptide levels could hold a pivotal role in assisting clinicians with the management of cardiovascular conditions, notably hypertension and heart failure.

Background: The objective of this evaluation was to determine the analytical and clinical performance of the AFIAS point-of-care (POC) Tn-I Plus assay (Boditech Med Inc).

Design And Methods: Limit of detection (LOD), limit of quantification (LOQ), repeatability, reproducibility, inter- and intra-individual CV were evaluated using the CLSI guidelines. The study was also designed to estimate the 99 percentile upper reference limit (URL) and to assess the diagnostic sensitivity and specificity.

Proteins represent powerful biomacromolecules due to their unique functionality and broad utility both in the cell and in non-biological applications. The genetic encoding of non-canonical amino acids (ncAAs) facilitates functional diversification of these already powerful proteins. Specifically, ncAAs have been demonstrated to provide unique functional handles to bioorthogonally introduce novel functionality via conjugation reactions.

  • Weill-Marchesani syndrome (WMS) is a genetic disorder marked by short stature, brachydactyly, joint limitations, and specific eye abnormalities like spherophakia and ectopia lentis. Cardiovascular issues can also occur. It has both dominant and recessive genetic forms caused by mutations in different genes.
  • A study involved 61 patients, including 18 from new research and 43 from existing literature, assessing the correlation between their genetic variations and clinical features. It was found that the majority exhibited eye anomalies and a significant portion had short stature and valvulopathy.
  • The research indicates that while eye abnormalities are crucial for diagnosing WMS, its overall symptoms appear more

Since the late 1990s, cathepsin K cleavage sites in type I collagen have been extensively studied due to its ability to release bone resorption biomarkers such as CTX and NTX. However, gel-based methods and N-sequencing used in these studies lack sensitivity, especially for small to medium peptides. In this work, we propose a degradomics mass spectrometry-based workflow that combines protein digestion, Nano-LC-UDMS, and several software tools to identify cathepsin K cleavage sites.

Background: Oxidative stress (OS) could cause various COVID-19 complications. Recently, we have developed the Pouvoir AntiOxydant Total (PAOT®) technology for reflecting the total antioxidant capacity (TAC) of biological samples. We aimed to investigate systemic oxidative stress status (OSS) and to evaluate the utility of PAOT® for assessing TAC during the recovery phase in critical COVID-19 patients in a rehabilitation facility.
