Here we present the Multisite Assembly of Gateway Induced Clones (MAGIC) system, which harnesses site-specific recombination-based cloning via Gateway technology for rapid, modular assembly of between 1 and 3 "Entry" vector components, all into a fourth, standard high copy "Destination" plasmid backbone. The MAGIC toolkit spans a range of in vitro and in vivo uses, from directing tunable gene expression, to driving simultaneous expression of microRNAs and fluorescent reporters, to enabling site-specific recombinase-dependent gene expression. All MAGIC system components are directly compatible with existing multisite gateway Tol2 systems currently used in zebrafish, as well as existing eukaryotic cell culture expression Destination plasmids, and available mammalian lentiviral and adenoviral Destination vectors, allowing rapid cross-species experimentation.

Systemic lupus erythematosus (SLE) is an autoimmune disease characterized by B-cell hyperactivity and breach of tolerance. Autoreactive memory B cells, which have a decreased activation threshold and the ability to survive in absence of antigen, are believed to contribute to chronicity in autoimmune diseases like SLE. Belimumab, the first approved biological treatment of active SLE and lupus nephritis, reduces B cells dependent on B-lymphocyte stimulator protein (BLyS) for survival, whereas memory B cells are spared; several studies reported circulating memory B-cell concentrations increase following BLyS neutralization.

Physiologically-based pharmacokinetic (PBPK) models are mechanistic models that are built based on an investigator's prior knowledge of the in vivo system of interest. Bayesian inference incorporates an investigator's prior knowledge of parameters while using the data to update this knowledge. As such, Bayesian tools are well-suited to infer PBPK model parameters using the strong prior knowledge available while quantifying the uncertainty on these parameters.

Stan is an open-source probabilistic programing language, primarily designed to do Bayesian data analysis. Its main inference algorithm is an adaptive Hamiltonian Monte Carlo sampler, supported by state-of-the-art gradient computation. Stan's strengths include efficient computation, an expressive language that offers a great deal of flexibility, and numerous diagnostics that allow modelers to check whether the inference is reliable.

Blue rubber bleb nevus syndrome (BRBNS) is a rare disease that presents as cutaneous and extra-cutaneous vascular malformations, most commonly affecting the gastrointestinal (GI) tract. We report a case of adult onset BRBNS in an African American male with vascular lesions isolated to the jejunum without any cutaneous manifestations. Physicians should recognize that BRBNS can present without skin involvement and may have complications from visceral organ involvement.

Peripheral neuropathy (PN) is a common long-term debilitating toxicity of antimicrotubule agents. PN was the most frequent adverse event resulting in dose modifications and/or discontinuation of treatment for valine-citrulline-monomethylauristatin E antibody-drug conjugates (ADCs) developed at Genentech. A pooled time-to-event analysis across eight ADCs (~700 patients) was performed to evaluate the relationship between the ADC exposure and the risk for developing a clinically significant (grade ≥ 2) PN.

A hazard model of fracture was developed using individual patient data (IPD) from the NHANES (2005-2008) database and summary-level data from an aggregate dataset (AD). The AD was built by performing a comprehensive and systematic literature search of clinical studies published from 1995 to 2015, recording fracture rate and bone mineral density (BMD) for both treatment and placebo arms. Spherical silver nanoparticles (NPs) of 30 nm diameter embedded in soda-lime glass were uniformly reshaped (elongated) after irradiation by a linearly polarised 250 fs pulsed laser operating within the NPs' surface plasmon resonance band. We observed second harmonic generation (SHG) and multiphoton-absorption-induced luminescence (MAIL) in the embedded laser-reshaped NPs upon picosecond (10 ps) pulsed laser excitation at 1064 nm. A complementary study of SHG and MAIL was conducted in soda-lime glass containing embedded, mechanically-reshaped silver NPs of a similar elongation ratio (aspect ratio) to the laser-reshaped NPs.

Radially and azimuthally polarized picosecond (~10 ps) pulsed laser irradiation at 532 nm wavelength led to the permanent reshaping of spherical silver nanoparticles (~30 - 40 nm in diameter) embedded in a thin layer of soda-lime glass. The observed peculiar shape modifications consist of a number of different orientations of nano-ellipsoids in the cross-section of each written line by laser. A Second Harmonic Generation cross-sectional scan method from silver nanoparticles in transmission geometry was adopted for characterization of the samples after laser modification.

A Potassium Titanyl Phosphate (KTP) crystal has been used in conjunction with a 10-ps pulsed laser to produce a frequency-doubled conically-refracted Gaussian beam. The 'free' and 'forced' beams that make up the scheme for nonlinear conical refraction were readily observable in non-phase-matched conditions. The dependency of the frequency-doubled beam patterns on the incident beam polarization, which until now has remained unexplored, was examined in detail.

A general description of electro-optic detection including non-collinear phase matching and finite transverse beam profiles is presented. It is shown theoretically and experimentally that non-collinear phase matching in ZnTe (and similar materials) produces an angular chirp in the χ(2)-generated optical signal. Due to this, in non-collinear THz and probe arrangements such as single-shot THz measurements or through accidental misalignment, measurement of an undistorted THz signal is critically dependent on having sufficient angular acceptance in the optical probe path.

Azimuthal and radial polarization states of light are used to produce conical diffraction (CD) from a KGd(WO4)2 crystal. The patterns produced in the ring plane in each case display marked differences than those seen when linearly polarized incident light is used, with the production of a splitting of the CD ring into two concentric rings of equal intensity. The free space evolution for each type of polarization state is also experimentally recorded and investigated.

Picosecond (~10 ps) pulsed laser irradiation at 532 nm led to the efficient and scalable fabrication of dichroic areas in glass with spherical silver nanoparticles of ~30 - 40 nm in diameter embedded in a surface layer of thickness ~20 μm. The observed dichroism is due to the uniform and permanent shape transformation of the nanoparticles - from spherical to spheroidal shapes - throughout the irradiated areas and along the laser polarization direction, paving the way for affordable manufacture of polarization-selective diffractive optical elements. The shape modification threshold and the dichroism as a result of Surface Plasmon Resonance band separation were identified.
